我国部分地区献血人群中单纯疱疹病毒感染流行情况

摘　　要：目的了解中国献血人群中单纯疱疹病毒(HSV)的流行率、病毒载量及抗体检出情况。方法构建的实时荧光定量PCR体系,检测来自重庆、洛阳、乌鲁木齐、绵阳和柳州等5个采供血机构3 794(人)份献血者血浆标本的HSV DNA,并测定DNA阳性标本的病毒载量;同时随机抽取其中292(人)份标本用ELISA法检测Ig G和Ig M抗体的感染情况。结果在本组献血者人群中,发现43份HSV DNA反应性标本,占1.13%(43/3 794),5个采供血机构的检出率分别为重庆1.24%(20/1611)、洛阳0(0/624)、乌鲁木齐3.63%(14/413)、绵阳1.88%(9/479)和柳州0(0/667);病毒载量(9.15×102-9.65×104)(中位数3.53×103)cp/m L。292份经ELISA检测的标本中,Ig G抗体阳性率16.44%(48/292),Ig M抗体阳性率4.45%(13/292),Ig G、Ig M抗体双阳性率2.40%(7/292)。结论目前HSV在我国献血人群中存在一定的流行率,但是检测出的HSV DNA阳性标本的病毒载量较低;是否需要对特殊受血者做血液HSV检测值得进一步探讨。Objective To investigate the prevalence of herpes simplex virus in Chinese blood donors,the viral load and the detection of the serological antibodies. Methods A quantitative real-time PCR system was established to detect 3,794 HSV DNA in plasma from five Chinese blood centers,from Chongqing,Luoyang,Urumqi,Mianyang and Liuzhou. The viral load of positive samples was measured. At the same time,292 randomly selected plasma from the five blood centers were tested with ELISA method to detect Ig G and Ig M infections. Results In this blood donor population,43 HSV DNA-reactive samples were found,accounting for 1. 13%（ 43 /3 794）. The detection rate in five blood centers were Chongqing 1. 24%（ 20 /1611）,Luoyang 0（ 0 /624）,Urumqi 3. 63%（ 14 /413）,Mianyang 1. 88%（ 9 /479）,Liuzhou 0（ 0 /667）,respectively. The viral load range was 9. 15 × 102- 9. 65 × 104 cp / m L. The median viral load was 3. 53 × 103 cp / m L. In 292 specimens by ELISA,Ig G antibody positive rate was 16. 44%（ 48 /292） and Ig M antibody positive rate was 4. 45%（ 13 /292）.2. 40%（ 7 /292） of specimens were seropositive for HSV Ig G and Ig M antibodies. Conclusion Currently,there is a certain prevalence of HSV in Chinese blood donors,but the viral load detected in HSV DNA positive specimens was lower. Whether or not the blood recipient needs to do a HSV detection is worth further exploration.
