摘 要:目的:探索体外利用CD45RA磁珠分选供者外周血干细胞(PBSCs)中CD45RA^+细胞(处女T淋巴细胞)的可行性;比较去除CD45RA^+细胞后的干细胞产物(CD45RA-depleted cells)体外抗肿瘤活性有无明显减弱。方法:CD45RA磁珠分选供者外周血干细胞中的CD45RA^+细胞;流式细胞仪检测分选纯度以及分选后干细胞产物的细胞成分;分选前后干细胞产物分别与K562/Jurkat肿瘤株共培养,CCK-8检测其体外抗肿瘤活性。结果:CD45RA磁珠分选供者PBSCs中CD45RA^+细胞有效率达95.41%(88.21%~98.18%),分选后干细胞产物主要以CD3+CD4+和CD3+CD8+细胞为主,CD45RA-depleted产物与PBSCs相比体外抗肿瘤效应无明显减低(P>0.05)。结论:利用CD45RA磁珠去除供者PBSCs中的处女T淋巴细胞是可行的,去除CD45RA^+细胞后的DBSCs产物体外仍具有抗肿瘤活性。Objective To explore the immunomagnetic depletion procedure of CD45RA^+cells(Nave T cells) in donor peripheral blood stem cells(PBSCs) by using CD45 RA microbeads and to compare the killing activity against leukemia cells of PBSCs and CD45RA-depleted cells in vitro. Methods CD45 RA microbeads were used to deplete CD45RA^+cells in PBSCs.Immunophenotype changes were analyzed by flow cytometry and CD45RA-depleted cells were co-cultured with K562 / Jurkatcells.Killing activity was detected by CCK-8 assay;Results The procedure for CD45 RA depletion eliminated 95.41% of CD45RA^+cells in PBSCs and CD3+CD4+and CD3+CD8+cells were most isolated. Compared with PBSCs,the ability against leukemia cells of CD45 RAdepleted cells were not decreased(P〉0.05)at effector / target(E / T) ratios range between 5 ∶1 to 40 ∶1.Conclusion The depletion of CD45 RA + naive T cells from donor PBSCs is feasible and the depleted products show killing activity to leukemia cells.
