重组hIL-10联合MTX对AA大鼠IL-1β、TNF-α含量变化和影像学变化研究

摘　　要：目的:以重组人白细胞介素-10(human interleukin-10,h IL-10)及其联合甲氨碟呤(MTX)作用佐剂性关节炎大鼠模型,检测血清IL-1β、TNF-α含量的变化情况,研究重组h IL-10联合MTX治疗佐剂性关节炎大鼠的效果。方法:AA大鼠造模后,用测量体重、足肿胀度、关节炎指数及组织病理学来评价造模是否成功,各药物处理AA大鼠模型后,用酶联免疫吸附试验(ELISA)检测大鼠血清IL-1β、TNF-α的含量。用放射学检查来评估AA大鼠后足关节损伤的程度。结果:各治疗组IL-1β、TNF-α含量比模型组水平明显下降,各治疗组与模型组比较有显著性差异(P<0.05),联合用药组对AA大鼠血清IL-1β、TNF-α有明显的抑制作用,影像学检查提示IL-1β、TNF-α水平与大鼠足骨关节的损伤程度成正相关,联合用药组治疗后骨侵蚀评分最低。结论:各治疗组对佐剂性关节炎大鼠血清中细胞因子IL-1β、TNF-α有抑制作用,这种抑制作用与骨关节影像学改变显著相关。联合用药组能有效降低AA大鼠的IL-1β、TNF-α水平,改善AA大鼠足关节损伤的治疗作用。Objective： Adjuvant arthritis（ AA） rats interfered with recombinant h IL-10,methotrexate（ MTX） and IL-10 plus MTX separately,we detected the changes of IL-1β and TNF-α in rat＇s serum. Methods： AA rats after interfered with recombinant h IL-10,MTX and IL-10 plus MTX. The levels of IL-1β,TNF-α in serum of rats were measured by ELISA method. with radiological examination to evaluate the degree of AA paw joint damage. Results： Compared to normal control group,the IL-1β,TNF-α of treatment group was significantly decreased,there was a significant differences compared with control group（ P〈0. 05）,The Combination group had significant difference compared with MTX group and IL-10 group,radiological examinations showed IL-1β,TNF-α levels and degree of AA paw joint damage were positive correlation,the combination group therapy bone erosion score lowest. Conclusion： Marked alteration in the serum level of IL-1β,TNF-α on Each treatment group of adjuvant arthritis in rats,these alterations of cytokine levels were associated with significant radiological changes of the joints. The combined group can effectively reduce levels of IL-1β,TNF-α and improve therapeutic effect of paw joint injury in AA rats.
