摘 要:目的:对双丹口服液(Shuang Dan oral liquid,SD)的入血成分进行分析研究,进而探讨其药效物质基础。方法:采用血清药物化学的研究方法,建立双丹冻干粉经口灌胃后大鼠血清的HPLC指纹图谱,结合体外样品指纹图谱,分析比较双丹冻干粉及单味药材给药后所得血清样品,鉴定双丹冻干粉大鼠血中移行成分。结果:通过与文献中标准物质的保留时间确定大鼠口服双丹冻干粉后有8种血清移行原型成分,分别为源自牡丹皮的没食子酸、芍药苷和丹皮酚,源自丹参的丹参素、原儿茶酸、咖啡酸、迷迭香酸和丹酚酸B,此8种成分将有可能成为双丹方药效作用的物质基础。结论:本实验初步确定了双丹口服液的入血成分,为后续双丹方体内直接作用物质的确定和药效相关性研究提供了数据支持,并为其药效物质基础的阐明奠定了理论基础。Objective: To Study on the therapeutic material bases of ShuangDun oral liquid (SD) by serum pharmacochemistry approach. Methods: HPLC fingerprints of dosed serum after oral administration of SD lyophilized powder were established. By comparative analysis of the chemical profiles of lyophilized powder, blank serum and dosed serum, potential bioactive compounds in SD could then be discovered. Results: After comparing the retention time with those of reference standard, 8 prototype compounds from SD lyophilized powder were discovered as potential bioactive components in rat serum. They are gallic acid, paeoniflorin and paeonol from Cortex Moutan and danshensu, protocatechuic acid, caffeic acid, rosmarinic acid and salvianolic acid B from Radix Salvia Miltiorrhizae. Conclusion: The constituents of SD absorbed into blood are preliminarily identified, which could provide fundament evidence for the therapeutic material bases of ShuangDan prescription.
