摘 要:目的在体外通过细胞培养,研究非甾体消炎药舒林酸对结肠癌HT-29细胞生长的影响及其引起细胞死亡的机制。方法采用四甲基偶氮唑蓝(MTT)比色法研究舒林酸对HT-29细胞增殖的影响;应用流式细胞仪(Flow Cytometry,FCM)对舒林酸作用下HT-29细胞调亡情况进行分析;在透射电镜下观察舒林酸作用HT-29细胞前后的形态变化。结果 MTT法显示舒林酸抑制HT-29细胞增殖呈时间、剂量依赖性;FCM法显示舒林酸促进细胞的凋亡;透射电镜显示,舒林酸作用后HT-29细胞后出现凋亡小体、染色质凝聚、核仁消失及核碎裂。结论舒林酸能抑制结肠癌HT-29细胞的生长,其机制可能与阻滞细胞周期的进程,促进细胞凋亡有关。Objective To study the effects and mechanisms of sulindac on the proliferation of the HT -29 cells and its apoptosis mechanisms with cell cuhure. Methods The proliferation effect of sulindac on HT - 29 cells was examined by MTT colorimetric assay. Inhibitory effects of apoptosis rates on HT - 29 cells were detected by flow cytometry before and after different concentratioixs of sulindac treated. Morphological changes of sulindac staining HT - 29 cells ,sere observed lay trans- mission electron microscopy. Results Sulindac inhibited the cells proliferation and induced apoptosis in a dose and time de- pendent manner. FCM showed that sulindac promoted apoptosis of the cells and sulindac - treated HT - 29 cells presented with apoptotic bodies, chromatin condensation, nucleolus and nuclear fragmentation. Conclusion Sulindac can inhibit the proliferation of HT - 29 cells, which may be related to its role in blocking cell cycle progression and induces cell apoptosis.
