小鼠肺癌原位模型的完善及三仙丸抑瘤作用初探

摘　　要：目的：进一步完善C57BL/6小鼠肺癌原位模型,并观察中药三仙丸对该模型小鼠肿瘤的抑制作用。方法：1在原有C57BL/6小鼠肺癌原位模型的基础上,根据造模原位接种C57BL/6小鼠肺腺癌细胞（Lewis lung cancer,LLC）和虫萤光素酶（luciferase）稳定结合的LLC-luciferase细胞株数量的不同,使用活体成像技术对模型小鼠活体成像曝光时间、肿瘤生长情况、肺癌的转移情况进行监测,并观察生存期,选出最优小鼠肺癌原位模型。2肺癌原位模型小鼠灌胃给服三仙丸,从造模第4天开始连续给药直至小鼠死亡,观察小鼠肿瘤大小和生存期。结果：活体成像时采用自动曝光效果最佳;造模接种细胞数≥1×10^5/只时小鼠肺癌原位模型稳定,且接种细胞数为1×10^5/只和5×10^5/只的模型小鼠中位生存时间分别为37.5 d和24.5 d（P〈0.05）。接种细胞数为1×10^5/只的小鼠,第39天时三仙丸组小鼠肿瘤明显小于对照组（P〈0.05）;对照组和三仙丸组小鼠中位生存时间分别为42 d和56 d（P〉0.05）。结论：构建小鼠肺癌原位模型时接种细胞数为1×10^5/只时造模效果佳;并用此模型证明了三仙丸对肿瘤的抑制作用。Objective： To further improve a C57 BL /6 mouse model of orthotopic lung cancer and observe the inhibitory effects of＂San Xian Pill＂on tumor growth in this optimized model. Methods： 1Luciferase-Lewis lung cancer（ LLC） cells were directly injected into the left lung of mice. Exposure time,tumor size and metastasis of lung cancer were observed by living imaging system,and the survival time was compared and analyzed to optimize the orthotopic model of lung cancer in mice. 2＂San Xian Pill＂was daily gavaged starting from the fourth day to death. Tumor size and survival time were observed.Results： When the number of LLC-luciferase cells was more than or equal to 1 × 10^5,the model of lung cancer in mice was stable. The median survival times were 37. 5 days and 24. 5 days for the 1 × 10^5 and 5 × 10^5 model mice respectively. In mice injected with 1 × 10^5LLC-luciferase cells,＂San Xian Pill＂significantly reduced the tumor size on day 39（ P〈0. 05）,and increased the median survival time from 42 days to 56 days. Conclusion： Mice injected with 1 × 10^5LLC-luciferase cells produced an optimal orthotopic lung cancer model,and＂San Xian Pill＂has inhibitory effects on tumor growth in this model.
