结直肠癌合并糖尿病患者组织中EMT相关蛋白的表达

摘　　要：目的观察结直肠癌合并糖尿病患者组织中促进上皮间质转换(EMT)进展的相关蛋白的表达。方法收集直肠癌及合并糖尿病患者的肿瘤组织及非肿瘤组织,分为对照组和肿瘤组;根据是否合并糖尿病,又分为伴及不伴糖尿病组。用免疫组化检测晚期糖基化终末产物(AGE)及其相应受体(RAGE)、上皮标志物钙黏连蛋白E(E-cad)、β-连环蛋白(β-catenin)、波形蛋白(vimentin);蛋白质印迹法检测各组中不同分化程度下相关蛋白的表达。结果 RAGE、AGE、E-cad和vimentin主要染色于细胞膜上,β-catenin主要染色于胞质内,且两个肿瘤组染色较对照组更深(P<0.01);与对照组相比,不伴糖尿病组及伴糖尿病中各蛋白表达均增高,E-cad表达降低(P<0.01);在同一分化状态下,伴糖尿病组中各蛋白表达水平较不伴糖尿病组高,E-cad较之降低(P<0.01)。结论结直肠癌合并糖尿病患者组织中相关蛋白β-catenin和vimentin表达上调,E-cad表达下降,从而促进EMT进展,对结直肠癌发生发展起重要促进作用。Objective To observe the expression of EMT-related protein in the tissue of colon rectum cancer（ CRC） patients with diabetes（ DB） in EMT progress. Methods Collect the tumor tissues of CRC patients combining with diabetes（ DB） and from controls. All the tissues were divided into control group and tumor group. The tumor group was divided into groups with DB and without DB. Associated protein RAGE,AGE,E-cad,β-catenin and vimentin were detected by immunohistochemical staining. The expression level of these protein were measured by Western blot. Results RAGE,AGE,E-cad and vimentin were mainly located in cell membrane,β-catenin was mainly located in cytoplasm,and the two tumor groups were stained deeper than control group（ P〈0. 01）. The expression level of AGE,RAGE,β-catenin and vimentin in two tumor groups were higher than those in control group,E-cad was lower（ P〈0. 01）. Compared with CRC without DB group,the expression level of AGE,RAGE,β-catenin and vimentin in the CRC with DB group were up-regulated,E-cad was lower than it in the same differential degree（ P〈0. 01）. Conclusions The relative protein β-catenin and vimentin are up-regulated while E-cad is down-regulated in the tissue of CRC patients with DB. The EMT progress is promoted which plays a vital role in the development of CRC.
