摘 要:目的观察红景天口服液对大鼠心肌细胞缺氧耐受性的改善作用。方法将40只雄性SD大鼠乳鼠,按体重随机分为溶剂对照组、模型对照组、阳性对照组和红景天组。溶剂对照组和模型对照组给予生理盐水20μL/m L,阳性对照组给予氟桂嗪胶囊20μg/m L,红景天组给予红景天口服液20μL/m L。大鼠心肌细胞提取后常规培养10 d,更换不含胎牛血清的DMEM培养基,溶剂对照组在正常环境的培养箱中培养;模型对照组、阳性对照组和红景天组心肌细胞放入温度为37℃及CO_2浓度为5%、O2浓度为1%的三气培养箱中进行缺氧处理,缺氧时间为6 h。缺氧后收集细胞培养上清液,按试剂盒的要求分别测试乳酸脱氢酶、丙二醛、肌酸激酶、超氧化物歧化酶的水平。取余下细胞,按凯基细胞凋亡检测试剂盒的要求用流式细胞仪测试细胞凋亡的情况,用MTT法测试心肌细胞的活力水平。结果红景天口服液能显著提高大鼠原代培养心肌细胞在缺氧条件下的活力,减少细胞凋亡,效果优于阳性对照药(P<0.01);并且可降低原代培养心肌细胞因缺氧所致心肌酶升高,有较好的清除氧自由基作用和对缺氧心肌细胞的保护作用,效果优于阳性对照药(P<0.01)。结论红景天口服液可明显提高大鼠心肌细胞的缺氧耐受性,为临床治疗缺氧性心肌病提供理论依据及实验资料。Objective To investigate the improvement of hypoxia tolerance of rat myocardial cells by Rhodiola rosea Oral Liquid.M ethods 40 male SD neonate rats were randomly distributed into 4 groups by their weights: solvent control group, model group, positive control group and the experimental group. The solvent group and the model group were treated with physiological saline( 20 μL / m L),while the positive control group was treated with physiological saline capsule( 20 μg / m L) and the experimental group with oral liquid of Rhodiola rosea( 20 μL / m L). The rat myocardial cells were cultured for 10 d in DMEM medium without fetal bovine serum. The solvent control group was cultured in incubator with normal conditions. The model group, positive control group and the experimental group were cultured in tri- gas incubat with 5% CO2 and 1% O2 at 37 ℃ for 6 h. Afterward, cell culture supernatant was harvested and analyzed through a number of assays. Based on the kit instruction, the levels of lactate dehydrogenase, malonaldehyde, creatine kinase, superoxide dismutase were subsequently measured. The rest cells were collected. The apoptosis of these cells was observed based on the Keki cell apoptosis detection kit instruction by flow cytometry, while the activity of these cardiac muscle cells was measured with the method of MTT. Results The whole experiment indicates that Rhodiola rosea Oral Liquid significantly increased the cell viability and reduced the cell death of rat primary cultured myocardial cells compared with positive control group( P〈0. 01). The assay performed indicated that the oral liquid of Rhodiola rosea lowered the myocardial enzymes level, which were induced by myocardial cell hypoxia and the levels of free radicals, showing that there was a better protective effect on hypoxic myocardial cells compared with positive control group( P〈0. 01). Conclusion Rhodiola rosea Oral Liquid can obviously improve the hypoxia tolerance of rat myocardial cells, and provide experimental data
