家蚕免疫血清体外抑制神经母细胞瘤细胞生长和增殖

摘　　要：家蚕免疫系统在抵抗病原微生物时会产生一些具有免疫功能的小分子肽,有研究表明这些肽具有抗肿瘤活性。我们通过免疫诱导法,给家蚕注射了一定量的抗原诱导家蚕产生免疫血清。从家蚕体内收集免疫血清,稀释成不同浓度梯度处理神经母细胞瘤细胞系BE(2)C,并观察其对BE(2)C细胞的影响。通过显微镜观察发现免疫血清处理后肿瘤细胞出现了大面积死亡。经MTT方法检测分析后发现,家蚕免疫血清对肿瘤细胞生长有抑制作用。实验结果初步证实了经抗原诱导的家蚕免疫血清对神经母细胞瘤系BE(2)C的生长及增殖有抑制作用,推测家蚕免疫血清可能诱导了肿瘤细胞发生凋亡,为进一步解析其调控机制提供了有力的数据支撑,为开发家蚕新的经济用途奠定了良好基础。Silkworm immune system produce some immuno-competent micromolecule peptide when they are resisted by microbial pathogens. Recently researches indicated that those micromol- ecule peptide had antibiotic and antitumor activities.. We inject a certain amount of antigen to induce silkworms produce immune serum. We collect the immune serum from the injected silkworm, and dilute it into different concentration gradient to treat neuroblastoma cell line BE（2）C, and observe the effect of the serum on BE（2）C cells. By microscopic observation, we found that there is a large amount of cells dead after the treatment of immune serum. De- tected by MTT analysis, the silkworm immune serum has inhibitory effect on tumor cell growth. The experimental results confirmed that the antigen induced silkworm immune ser- um inhibited the growth and proliferation of neuroblastoma cell line BE（2）C, speculated that the silkworm immune serum could induce apoptosis of tumor cells, provides a powerful data support for further analysis of the regulation mechanism, which lay a good foundation in de- veloping a new economic purposes of silkworm.
