摘 要:为检测我国异地小群保种地方鸭群体的遗传结构,利用荧光STR分型技术结合ABI-3730XL型DNA测序平台,对异地小群保种的9个品种在12个位点进行基因分型,利用SPSS19.0对所有等位基因行PCA分析,并使用Structure2.0软件进行群体结构分析。结果显示:12个位点的多态性丰富,共检测到90个等位基因,PCA分析得到8个主成分,前6个主成分对群体分类的累积贡献率达到88.091%。Structure构建的9个群体的遗传分化结构图显示靖西麻鸭与山麻鸭亲缘关系最远,攸县麻鸭与临武鸭距离最近,与PCA分析构建的三维散点图基本一致。In this study, the allele frequency of 12 microsatellite loci in 9 allopatry domestic duck groups were detected by STR genotyping method based on the ABI-3730XL DNA sequencing platform. The data were analyzed by SPSS19.0 and Structure2.0 software. The results showed that the polymorphisms of 12 loci were abundant and 90 alleles were obtained. The distributions of the alleles were analyzed by PCA and 8 principal components were got. The accumulating contribution rate of the first 6 ones was 88.091%. The genetic differentiation structure of the populations were structured by structure procedure. The genetic relationship between Jingxi duck and Shanma duck was the farthest, meanwhile that between Youxian and Linwu was the nearest. The results were consistent with the results of PCA analysis.
