壮骨止痛方对去卵巢骨质疏松大鼠血清瘦素及长形受体的影响  

Effects of Zhuanggu Zhitong Prescription(壮骨止痛方) on Serum Leptin and its Length Receptor in Ovariectomized Rats with Osteoporosis

摘　　要：目的:观察壮骨止痛方对去卵巢大鼠血清瘦素(leptin)及长形受体(OB-Rb)的影响,探讨其抗骨质疏松机制。方法:72只SD大鼠按体质量随机抽出假手术组12只,造模组60只,造模组采用双侧去卵巢法造模,假手术组只切除卵巢周围相应质量的脂肪。造模成功后,造模组大鼠按体质量随机分为模型组、壮骨止痛方高剂量组(13.2 g/kg)、中剂量组(6.6 g/kg)、低剂量组(3.3 g/kg)和戊酸雌二醇组,每组12只。术后第5天开始药物干预,模型组和假手术组给予相应体积的纯净水,持续13周。给药结束后,检测大鼠骨密度(BMD)和血清瘦素及其长形受体含量。结果:模型组大鼠血清OB-Rb含量及BMD均显著低于假手术组,差异均有统计学意义(P<0.05或P<0.01)。壮骨止痛方高、中、剂量组大鼠瘦素长形受体含量及BMD均显著高于模型组,差异均有统计学意义(P<0.05或P<0.01)。结论:壮骨止痛方具有促进去卵巢骨质疏松大鼠瘦素受体表达,增强瘦素的作用,这可能是其抗骨质疏松的作用机制之一。Objective：To explore the effects of Zhuanggu Zhitong Prescription （壮骨止痛方, ZGZTP） on serum Leptin and its Length Receptor （OB-Rb） in ovariectomized rats with Osteoporosis and investigate its mechanism in anti-osteoporosis. Methods： The 72 rats were randomly divided into sham group （12） and model group （60）, the model group rats were ovariectomized and the sham group rats were excised equal weight fat. After the modeling, the rats were randomly divided into model group, ZGZTP high-dose group （13.2 g/kg）, ZGZTP middle-dose group （6.6 g/kg）, ZGZTP low-dose group （3.3 g/kg） and estradiol valerate group, 12 rats in each group. 5 days after the operation, drug therapy started for 13 weeks, and the model group and sham group rats were given equal volume water. After the test, bone mineral density （BMD） and the content of leptin and its length receptor in the serum were detected. Results： The content of serum OB-Rb and the BMD in model group was significantly lower than the sham group, the difference was statistically significant （P〈0.05 or P〈0.01）. The contents of serum OB-Rb and the BMD in ZGZTP high-dose, middle-dose and low-dose group were higher than the model group, and the difference was statistically significant （P〈0.05 or P〈0.01）. Conclusion： ZGZTP has the effect of promoting the expression of leptin receptor in ovariectomized osteoporosis rats and enhancing the role of leptin, and this may be one of the mechanisms of its anti osteoporosis.
