摘 要:目的探讨二烯丙基硫化物(DAS)对百草枯(PQ)中毒大鼠急性肺损伤的影响。方法 100只雄性Wistar大鼠随机分为5组,正常对照组、PQ 70 mg·kg^(-1)模型组、DAS 25,50和100 mg·kg^(-1)治疗组,每组20只。一次性ig给予PQ 70 mg·kg^(-1)制备PQ中毒模型,DAS组于造模前、后30 min ip给予相应剂量药物,正常对照组ip给予等量生理盐水。各组分别于造模后1,3,6和12 h麻醉大鼠,取右肺下叶进行HE染色,光镜下观察肺组织病理损伤程度并评分;右肺上叶进行肺组织一氧化氮(NO)含量检测;左肺进行支气管肺泡灌洗获取肺泡巨噬细胞(AM),培养24 h取上清,分光光度法测定诱导型一氧化氮合酶(i NOS)含量;培养至72 h,采用细胞免疫化学方法检测AM中i NOS蛋白的表达。结果与正常对照组比较,模型组肺泡结构破坏严重,病理评分明显升高(P<0.01),肺组织中NO含量明显升高(P<0.01),AM培养上清液中i NOS含量明显升高(P<0.01),i NOS蛋白表达增加(P<0.05);与模型组比较,DAS 50 mg·kg^(-1)组在造模后3,6和12 h以及DAS 100 mg·kg^(-1)组在造模后1,3,6和12 h肺泡结构损伤显著减轻,病理评分降低(P<0.05);DAS25和50 mg·kg^(-1)组肺组织中NO和i NOS含量及i NOS蛋白表达减少(P<0.05),DAS 100 mg·kg^(-1)组NO含量减少更为明显(P<0.01)。结论 DAS可能通过抑制PQ中毒大鼠肺组织中的氧化损伤因子减轻肺损伤。OBJECTIVE To investigate the effect of diallyl sulfide(DAS) in protection against acute lung injury in rats induced by paraquat(PQ). METHODS A total of 100 male Wistar rats were randomly divided into normal control group,PQ 70 mg·kg^-1model group,and DAS 25,50 and 100 mg·kg^-1treatment groups,with 20 rats in each group. A poisoning model was estalolished after administration ig at a single dose of PQ 70 mg·kg^-1,while the normal control group was ip given the same volume of normal saline. DAS 25,50 and 100 mg·kg^-1was intraperitoneal y injected 30 min before and after PQ exposure.Five rats in each group were sacrificed at 1,3,6 and 12 h,respectively. The inferior lobe of the right lung was observed by HE staining under an optical microscope. Tissue of the upper lobe of the right lung was used to detect the content of nitric oxide(NO). Alveolar macrophages(AMs) were collected and cultured for 24 h,and the content of nitric oxide synthase(i NOS)in the supernatant was detected. AMs were cultured for 72 h and the expression of i NOS protein in AMs was detected by immunocytochemistry method. RESULTS Compared with normal control group,the alveolar structure of PQ group was severely damaged and the pathological score was significantly increased(P〈0.01).The NO content of PQ group was significantly higher than in normal control group(P〈0.01). The content and protein expression of i NOS were significantly increased in PQ group(P〈0.01). Compared with PQ group,the lung injury score of rats in DAS 50 mg·kg^-1group at 3,6 and 12 h and in the DAS 100 mg·kg^-1group at each time point was decreased(P〈0.05). Compared with PQ group,the NO content of DAS25 and 50 mg·kg^-1group was decreased(P〈0.05),and the NO content of DAS 100 mg·kg^-1group was significantly reduced(P〈0.01). The content of i NOS was reduced in DAS 100 mg·kg^-1group(P〈0.05). Compared with PQ group,the expression of i NOS protein in DAS groups was decreased(P〈0.05). CONCLUSION DAS can inhibi
