摘 要:菜豆普通花叶病毒(BCMV)、菜豆黄花叶病毒(BYMV)和南方菜豆花叶病毒(SBMV)是黑龙江省菜豆主要的种子传播病毒。为分析菜豆种质资源携带病毒的状况,以6份普通菜豆种子的胚根、胚芽和子叶为试材,采用CTAB法提取总RNA和基因组DNA,根据BCMV、BYMV和SBMV的衣壳蛋白基因序列设计特异引物,通过PCR和RT-PCR对180份菜豆资源进行病毒检测。结果表明:RT-PCR对BCMV、BYMV和SBMV的检出率分别为12.78%、9.44%和1.67%;PCR的检出率分别为12.78%、8.33%和1.67%。所收集的6种菜豆种质资源均检出1~3种病毒,其中BCMV和BYMV是主要的病毒种类。Bean common mosaic potyvirus(BCMV),bean yellow mosaic virus(BYMV)and southern bean mosaic virus(SBMV)are the important seed borne pathogen of Heilongjiang common bean.For analyzing the status of common bean germplasm,taking the radicles,plumules,and cotyledons of six common bean seeds as materials,the genomic DNA and total RNA was extracted from different organs by CTAB method.Specific primers were designed according to published coat protein(CP)gene sequence of BCMV,BYMV and SBMV,respectively.180 germinated seeds of common bean were tested for the presence of the three viruses by polymerase chain reaction(PCR)and reverse transcription-polymerase chain reaction(RT-PCR).The results of RT-PCR revealed that the average infection frequencies of BCMV,BYMV,and SBMV in common bean samples were 12.78%,9.44%and 1.67%,respectively.Results of PCR revealed that the average infection frequencies of BCMV,BYMV and SBMV in common bean samples were 12.78%,8.33% and 1.67%,respectively.All six common bean varieties tested were positive one,two or three viruses.The causal viruses of common bean were BCMV and BYMV.
