摘 要:实时荧光定量PCR技术将光谱技术融合于传统PCR技术上,通过荧光信号积累实时监测PCR进程,实现核酸定量,具有特异性强、敏感性高、定量精准、自动化程度高等优点,被广泛运用于分子生物学等领域。本文结合近几年的研究成果,对实时荧光定量PCR技术的原理、检测方法、引物设计以及在家禽遗传育种的应用和存在的问题等进行了综述。Real-time fluorescent quantitative PCR is a kind of technology that combines spectroscopy with traditional PCR and the nucleic acid quantitative can be realized through the real-time PCR monitoring of fluorescent signal accumulation,with the advantages of specificity,good sensitivity,precise quantitative and high automation. This technique is widely applied in such fields as molecular biology. Based on the recent achievements of research,the principles,the detection methods,the primer design of PCR and its present application to poultry genetics and breeding and the existing problems are all reviewed in this paper.
