摘 要:目的:探讨Toll样受体4(TLR4)基因Asp299Gly位点、Thr399Ile位点多态性与肺结核及结核性胸膜炎的遗传易感性。方法:采用错配聚合酶链反应—限制性片段长度多态性分析(PCR-RFLP)及PCR-直接测序法在190例结核患者(其中结核性胸膜炎患者64例)和110例健康自愿者人群(对照组)中检测TLR4基因Asp299Gly位点和Thr399Ile位点的突变频率,并对实验结果进行统计分析。结果:在结核组、结核性胸膜炎组和对照组中均未发现TLR4基因Asp299Gly位点和Thr399Ile位点的突变。结论:TLR4基因Asp299Gly位点和Thr399Ile位点多态性与肺结核及结核性胸膜炎的遗传易感性无关。Objective:To investigate the association between the Asp299 Gly and Thr399 Ile polymorphisms of the Toll-like receptor 4(TLR4)gene and susceptibility to pulmonary tuberculosis,and/or tuberculous.Methods:TLR4gene Asp299 Gly allele and Thr399 Ile allele mutation were screened by polymerase chain reaction-restricted fragment length polymorphism(PCR-RFLP)analysis technique in 190 pulmonary tuberculosis patients,including 68 tuberculous pleuritis patients and 110 healthy volunteers.Results:No mutation of TLR4 gene Asp299Gly allele and Thr399 Ile allele were detected in all 300 DNA samples.Conclusion:The non-synonymous single nucleotide polymorphism Asp299 Gly and Thr399 Ile in TLR4 gene was rare in Chinese population in Guangxi province,and had no association with the susceptibility to tuberculosis and/or tuberculous pleuritis.
