摘 要:目的:探讨芍药苷对原代培养的大鼠心肌细胞缺氧/复氧损伤的保护作用.方法:体外培养原代大鼠心肌细胞,建立缺氧/复氧损伤模型,MTT法观察芍药苷对缺氧复氧乳鼠心肌细胞存活及培养液LDH活力的影响;流式细胞仪观察芍药苷对缺氧复氧乳鼠心肌细胞凋亡的影响;荧光定量PCR法观察芍药苷对缺氧复氧乳鼠心肌细胞Caspase-3 mRNA表达的影响.结果:与模型损伤组比较,与缺氧模型组比较,芍药苷可抑制缺氧引起的心肌细胞损伤,减少LDH的漏出(P<0.05);明显抑制心肌细胞缺氧复氧所致的早期凋亡(P<0.01);明显抑制缺氧复氧后心肌细胞Caspase-3 mRNA的表达.结论:芍药苷对原代培养大鼠心肌细胞缺氧/复氧损伤的保护作用机制可能与其减少缺氧引起的细胞损伤、通过caspase-3蛋白抑制缺氧/复氧心肌细胞早期凋亡有关.OBJECTIVE : To explore the effect of Shaoyaogan on hypoxia and reoxygenation injury of cardiac muscle cell in mice. METHODS : Cardiac muscle ceils of the mice were cultured in vitro and the modle of hypoxia and reoxygenation injury was mede. The survive of cardiac muscle cells and the activity of LDH were observed. The death of cardiac muscle cells of those mice was observed by fluid cell equipment. The express of Caspase - 3 mRNA of these cells of mice was observed by PCR. RESULTS : In hypoxia group Shaoyaogan could inhibit the injury of cardiac muscle cell caused by hypoxia and reduce leakege of LDH compared with injury group (P 〈0. 05 -0. 01); It could inhibit early death of cardiac muscle cell caused by hypoxia and reoxygenation (P 〈0. 01 ); And it might inhibit the express of easpase -3 mRNA of cardiac muscle cell after reoxygenation signifieanly. CONCLUSION: Protective machanism of the injury of cardiac muscle cell caused by hypoxia and reoxygenation are related to cell injury caused by hypoxia and the early death of cardiac muscle cells of hypoxia and reoxygenation inhibited by caslaase - 3.
