摘 要:目的:研究黄芪预处理对大鼠肠系膜缺血再灌注损伤的保护作用。方法:取120只实验用大鼠随机分假手术组、模型组、银杏叶提取物(3.15 mg/kg)预处理组和黄芪(5、10和20 g/kg)预处理组;术前7天开始腹腔注射给药(1次/d),假手术组和模型组给予等体积生理盐水;通过夹闭肠系膜上动脉45 min后去夹恢复血流再灌注的方法制备肠系膜缺血再灌注损伤大鼠模型;再灌注2 h后,取肠组织观察并测定肠组织含水量,HE染色观察肠组织病理变化并进行损伤评分(Chiu's氏评分);检测肠组织中抗氧化酶(SOD、CAT)活性和丙二醛(MDA)含量;酶联免疫法(ELISA)测定肠组织中炎症细胞因子(CRP、TNF-α、IL-1β、IL-6、IL-10)含量;末端标记法(TUNEL)检测肠系膜细胞凋亡状况并计算凋亡指数(Apoptosis Index,AI),通过比色法定量分析肠组织中Caspase-3、Caspase-9活性,Western blot法测定肠组织中NF-κB蛋白表达并进行半定量分析。结果:与模型组比较,黄芪预处理组大鼠肠组织外观明显红润、水肿状况明显改善,其中10、20 g/kg预处理组肠组织含水量显著降低(P<0.05,P<0.01);黄芪预处理各组大鼠肠组织病变明显较轻,其中10、20 g/kg预处理组Chiu's氏评分显著低于模型组(P<0.05、P<0.01);黄芪(10、20 g/kg)预处理组肠组织中SOD、CAT活性显著升高且MDA含量显著降低(P<0.05,P<0.01),CRP、TNF-α、IL-6含量显著降低(P<0.05,P<0.01),20 g/kg预处理组IL-1β含量显著降低且IL-10含量显著升高(P<0.05);黄芪预处理各组肠系膜细胞凋亡状况明显较轻,其中10、20 g/kg预处理组肠系膜细胞AI显著低于模型组(P<0.01),肠组织中Caspase-3、Caspase-9活性显著降低、NF-κB蛋白表达显著下调(P<0.05,P<0.01)。结论:黄芪预处理对大鼠肠系膜缺血再灌注损伤具有保护作用;作用机制可能与黄芪能够改善抗氧化酶活性、提高自由基清除能力,抑制炎症细胞因子表达,降低Caspase-3、Caspase-9活性,抑制NFObjective: To investigate the protective effects of Astragalus on intestinal of ischemia- reperfusion rats. Methods: 120 experimental rats were randomly devided into six groups: sham operation group,model group,Ginkgo biloba extract( GB,3. 15 mg / kg) pretreatment group and Astragalus( 5,10,20 g / kg) pretreatment groups. Severn days before surgery,the drugs were given by intraperitoneal injection,once a day.The rat models were induced by clipping superior mesenteric artery; two hours later,the appearance of intestinal tissue was observed and the water content was detected; the histopathological changes of intestinal tissue was observed by HE staining and the Chiu's score was detected; the activity of SOD,CAT and the content of MDA in intestinal tissue were detected; the level of CRP,TNF- α,IL- 1β,IL- 6,IL- 10 in intestinal tissue were detected; the apoptosis of intestinal cells was observed by TUNEL staining and the apoptosis index( AI) was calculated; the activity of Caspase- 3,Caspase- 9 in intestinal tissue was detected; the expression of NF- κB was detected by Western blot. Results: Compared with the model group,the appearance of intestinal tissue in astragalus pretreatment groups was improved,and the water content in Astragalus( 10,20 g / kg)pretreatement groups was significantly decreased( P 〈0. 05,P〈 0. 01); the histopathological changes of intestinal tissue in Astragalus pretreatement groups was significantly improved,and the Chiu' s score in Astragalus( 10,20 g /kg) pretreatement groups was significantly decreased( P〈 0. 05,P 〈0. 01); the activity of SOD,CAT in intestinal tissue was significantly increased and the content of MDA was significantly decreased( P 〈0. 05,P 0. 01); the level of CRP,TNF- α,IL- 6 in intestinal tissue of Astragalus( 10,20 g / kg) pretreatement groups was significantly decreased( P 〈0. 05,P 〈0. 01),and the level of IL- 1β in 20 g / kg pretreatement group was significantly decreased and the IL- 10 was significa
