Imp-L3表达量降低与规律运动对老龄果蝇心脏功能的影响

摘  要:目的研究老年时期规律运动和蜕皮素诱导基因表达量降低对老龄果蝇心脏功能的影响,同时对果蝇老年时期运动抗心脏衰老的效果进行评价。方法安静组、干扰组、运动组和干扰运动组前6周均保存在标准培养箱内,第7周开始运动时,保存在30℃、其他条件与之前相同的培养箱内。第44天龄开始,运动组和干扰运动组果蝇采用运动训练平台装置进行规律性训练,环境温度为30℃,每天运动2.5 h,连续运动5天。全部运动结束后的第2天解剖果蝇,利用M-mode测试系统检测其心功能。结果运动结束后,收缩直径安静组小于干扰组(P<0.05),运动组小于干扰运动组(P<0.05);舒张直径安静组小于运动组(P<0.01);纤维性震颤安静组低于干扰组(P<0.05),干扰运动组低于干扰组(P<0.05),运动组低于安静组(P<0.05);射血分数安静组高于干扰组(P<0.01),干扰运动组高于干扰组(P<0.01)。结论蜕皮素诱导基因表达量的降低使果蝇心脏收缩力减弱,纤维性震颤的发生率升高,射血分数减少,而运动能够减少老龄果蝇心脏纤维性震颤的发生率,增加射血分数,有效提高果蝇心肌收缩力。O bjective Study the effect of regular exercise and decreased expression of ecdysone-inducible gene L3 on the cardiac function of ageing drosophila melanogaster,and assess the efficacy of senescent heart which is exercised regularly with age. Method: control group、UAS-Imp-L3 RNAi group、exercise group、UAS-Imp-L3 RNAi exercise group were conserved in 18℃ standard incubator( constant temperature and 50% humidity,12 /12 hours a day and night light cycle)among first six weeks. Starting from the seventh week,they were shifted to 30℃ standard incubator. Began with 44-day-old,flies that exercise group and UAS-Imp-L3 RNAi exercise groups were trained regularly at sports training platform in a constant 30℃ temperature room,the training sustain 5 days,2. 5h each day. Finished the exercise procedure,dissected drosophila melanogaster and detected its cardiac function with M-mode test system. R esults The systolic diameters of control group is lower than UAS-Imp-L3 RNAi group( P 0. 05),and exercise group is lower than UAS-Imp-L3 RNAi exercise group( P 0. 05). The diastolic diameters of control group is lower than exercise group( P 0. 01). Fibrillation of control group is lower than UAS-Imp-L3 RNAi control group( P 0. 05),fibrillation of control group and UAS-Imp-L3 RNAi control group were respectively higher than exercise group( P 0. 05) and UAS-Imp-L3 RNAi exercise group( P 0. 05). Ejection fraction of UAS-Imp-L3 RNAi exercise group was higher than UAS-Imp-L3 RNAi control group( P 0. 01). C onclusion The decreased expression of ecdysone-inducible gene L3 can reduce drosophila melanogaster's cardiac contractility and ejection fraction,and make its incidence rate of fibrillation heightened. While exercising regularly can reduce the incidence rate of fibrillation of drosophila melanogaster,and can increase its ejection fraction,efficiently improve its cardiac contractility.
