摘 要:为选育兼具较强的生根能力和抗逆性的转基因三倍体毛白杨,以3个rolB+rolC拷贝数为1+1型转基因毛白杨无性系、6个2+1型rolB+rolC转基因毛白杨无性系和4个1+2型rolB+rolC转基因毛白杨无性系为研究对象,观察和分析了其叶片解剖结构。结果表明:2+1型、1+2型rolB+rolC转基因三倍体毛白杨无性系的变异方向和变异程度未呈现一定规律,但与rolB+rolC拷贝数为1+1型的转基因毛白杨无性系相比,无性系301B、263B、363B及163C的紧密度均显著增加,无性系101C紧密度增加,而其它无性系的紧密度则显著减小。本研究可为选育兼具生根能力及抗逆性的优良三倍体毛白杨提供参考。In order to breed transgenic plants with strong rooting ability and resistance to stress,we studied the anatomical structure of triploid poplar leaves.Three clones with one rolB and one rolCgene,six clones with two rolBand one rolCgene,and four clones with one rolB and two rolCgenes of transgenic triploid Populus tomentosatissue culture seedlings were used as materials.The results showed that the transgenic clones with different copies of rolBand rolCgenes did not perform certain regularities in direction and degree of variation.However,compared with triploid Populus tomentosa with one rolBand one rolCgene,the tightness of leaf palisade tissue structure of clone 101 Cincreased,and that of the clone 301 B,263B,363Band163 Cincreased significantly,while the other clones decreased significantly.This study can provide a reference for the selection of both rooting ability and excellent resistance of transgenic triploid Populus tomentosa.
