摘  要:目的从富含多酚和多糖的狭叶红景天的茎和叶中提取出高质量RNA。方法比较了两种RNA提取方法,发现Trizal试剂法提取的RNA效果更好;同时,建立了一套完整检测RNA质量的科学方法。结果应用该方法从狭叶红景天植物茎提取的RNA的纯度是:A260/A280为2.098,A260/A230为1.138,浓度为214 ng/μl,电泳条带清晰,RNA完整性好。由此法所确定的高质量RNA可直接用于分子生物学实验。结论该方法可有效地从富含多酚和多糖的狭叶红景天的茎中提取高质量RNA,为高原珍稀药用植物分子生物学研究奠定基础。Objective To explore the high quality total RNA isolation method from stems and leaves of Rhodiola kirilowii Maxim.containing much polyphenols and amyloses. Methods Rapider procedure for high- quality total RNA isolation was obtained comparing two RNA isolation methods. At the same time,we established a set of complete and scientific method for testing the quality of RNA. Results A260/ A280 and A260/ A230 of prepared RNA were respectively 2. 098 and 1. 138,and RNA concentration was 214 ng / μL. The result of electrophoresis showed that the RNA bands of stem which was delt by trizal were clear and showed no degradation. This high quality RNA could meet the demands of molecular analysis which was ensured by our method. Conclusion This method is able to explore the high quality total RNA solation method from stems of Rhodiola kirilowii Maxim. containing much polyphenols and amyloses,and there will be a foundation for the study of plantwhich is regarded as rare medicine by molecular biology.
