摘 要:目的:建立T〉MIC的简易数学模型,通过%T〉MIC理论指导哌拉西林他唑巴坦给药方案的设计与优化。方法:以一室药动学模型为基础,建立等间隔等剂量重复静滴给药的T〉MIC的数学模型,依据该模型结合哌拉西林他唑巴坦对临床常见感染细菌的敏感性折点计算%T〉MIC值,以%T〉MIC达到50%100%为目标阈值对不同的细菌感染设计和优化给药方案。结果:建立了一种计算简便的T〉MIC数学模型,通过该模型计算的%T〉MIC可初评与优化临床常用方案。结论:本模型用于哌拉西林他唑巴坦抗菌方案的初设与优化方便、快捷。OBJECTIVE To establish a simple mathematical model of times above MIC(T〉MIC),guide theoretic design and optimization of dosing regimens of piperacillin/tazobactam by ratio of plasma concentration above MIC to dosing interval(%T〉MIC).METHODS Based on one compartment pharmacokinetic model,a simple mathematical model of T〉MICwas established by repeated intravenous administration at the same dose every time within the same interval,and used to calculate %T〉MIC of dosing regimens of piperacillin/tazobactam by sensitivity breakpoints against clinically infective bacteria.Range of 50% to 100% of %T〉MIC was used as target threshold to design and optimize dosing regimens against different bacterial infections.RESULTS The simple mathematical model of T〉MICbased on %T〉MIC was easily calculated and%T〉MICcould be used to evaluate and optimize dosing regimen roughly.CONCLUSION The model for calculation of T〉MICis easy and quick,can be used for preliminary design and optimization of piperacillin/tazobactam antibacterial regimens.
