摘　　要：目的观察MAGE-A4 mRNA在脑胶质瘤中的表达情况,探讨其表达与病理级别、KPS评分等临床参数及总生存期的关系。方法采用逆转录聚合酶链反应(RT-PCR)方法检测MAGE-A4 mRNA在78例脑胶质瘤组织、12例正常脑组织和7例正常睾丸组织中的表达,分析其表达与性别、年龄、KPS评分及病理级别等临床参数及总生存期关系。结果 MAGE-A4 mRNA在正常睾丸组织、正常脑组织及胶质瘤中的表达率分别为100%、0%、47.44%,差异有统计学意义(P<0.05);MAGE-A4 mRNA表达与病理分级相关(P<0.05);胶质瘤患者总生存期与病理分级、KPS评分相关(P<0.05)。结论 MAGE-A4在脑胶质瘤组织中有特异性表达,并且与病理级别显著相关,而在正常脑组织中没有表达,故MAGE-A4可以作为脑胶质瘤免疫治疗的理想靶点;胶质瘤患者KPS评分和病理分级,可作为判断患者预后依据。Objective To study aims to detect the expression of MAGE-A4 mRNA in glioma tissues, and then analyze the relationship among MAGE-A4 mRNA expression, clinical parameters and overall survival of patients with glioma. Methods RT-PCR （reverse transcription polymerase chain reaction） was adopted to investigate the expression of MAGE-A4 mRNA in 78 glioma tissues, normal brain tissues and normal testis tissues. The correlation between MAGE-A4 mRNA expression and clinical parameters like gender, age, KPS score and pathological grade of glioma patients was analyzed. Rseults MAGE-A4 positive rates in normal testis tissues, normal brain tissues and glioma tissues were 100%, 0% and 47.44%, respectively. The difference was statistically significant （P〈0.05）. A statistically significant difference was observed between the MAGE-A4 expression and pathological grade （P〈0.05）. Overall survival of glioma patients was significantly correlated with pathological grades and KPS score （P〈0.05）. Conclusion MAGE-A4 is expressed in glioma tissues but not in normal brain tissue, and the expression is significantly correlated with glioma pathological grade, therefore, MAGE-A4 may be ideal target for glioma immunotherapy. KPS score and pathological grade may be considered as an indicator of glioma prognosis.
