摘  要:针对大型沼气工程CO_2含量高、沼气利用率低及沼液中有机物含量高等问题,筛选得到能同时固定CO_2和去除沼液中有机物的光合细菌沼泽红假单胞菌(NJ),并对其进行条件试验。以模拟沼液固定化培养基为底物,考察光照强度、菌种的接种量和菌龄、培养基中Na2S的添加量等因素对固定CO_2速率的影响。以沼液为底物,考察沼液的稀释倍数和电子供体对光合细菌生长、固定CO_2速率和沼液化学需氧量(COD)去除率的影响。结果表明:以模拟沼液为培养基时,在温度35℃、光照强度1 500 lx、接种量0.1 g/L、菌龄60 h及培养基中Na2S添加量2.0 g/L条件下,固定CO_2速率达205 m L/h(以1 g干细胞计)。初始沼液COD 3 g/L,将沼液稀释10倍,蔗糖添加量为0.15 g/L,Mg CL2添加量为0.06 g/L的条件下,光合细菌NJ生长最快,光密度(OD)值由0.24增大到0.48;固定CO_2速率达到最大,为218 m L/h。此时,沼液COD去除率为93%。Screened photosynthetic bacteria Rhodopseudoumonas palustris( NJ) was used for fixing carbon dioxide and removing organic matter in biogas slurry and the condition test was studied to solve problems in large-scale biogas project,such as high CO_2 content,low biogas utilization,high organic content were studied. Immobilized substrate was used as analog biogas slurry to investigate the factors effecting CO_2 fixation rate such as the light intensity,bacteria inoculation amount and ages,medium addition of Na2 S.Biogas dilution factor and the added electron donor for CO_2 fixation rate and slurry COD removal rate were further investigated using biogas slurry as substrate. Results showed as follows: when analog biogas slurry as substrate,at temperature of 35 ℃,light intensity of 1 500 lx,inoculum amount of 0. 1 g / L,bateria age of 60 h and Na2 S amount of 2. 0 g / L,CO_2 fixed rate was up to 205 m L / h( in 1 g dry cell). Under conditions of initial biogas slurry COD of 3 g / L,dilution of 10 times,sucrose of 0. 15 g / L and Mg Cl2of0. 06 g / L,NJ could obtain the fastest growing speed,optical density( OD) value increased from 0. 24 to0. 48,and CO_2 fixation rate reached the maximum of 218 m L / h. Meanwhile,biogas slurry COD removalrate was 93%.
