摘  要:研究川芎嗪与黄芪甲苷不同配伍浓度对鸡胚绒毛尿囊膜模型(chick embryochorioallantoic membrane,CAM)血管新生的作用,探讨二药配伍对缺血性心脏病的影响.将100只8 d龄鸡胚随机分为10组,包括空白DMSO对照组、不同浓度的川芎嗪(1、2、4 mg/m L)与黄芪甲苷(0.2、0.4、0.8 mg/m L)两两配伍组共9组,每组10只鸡胚.采用鸡胚绒毛尿囊膜(CAM)法,通过混合纤维素微孔滤膜载体将药物加到CAM表面上,继续孵化3 d观察血管生长状况,并进行新生血管计数.结果表明,川芎嗪与黄芪甲苷各浓度配伍均能促进鸡胚绒毛尿囊膜血管新生,且与浓度有一定的剂量依赖关系.其中,川芎嗪2 mg/m L+黄芪甲苷0.4 mg/m L一级血管与二级血管管与空白组比较,有明显差异(P<0.001,P<0.001).川芎嗪黄芪甲苷配伍可促进鸡胚绒毛脲囊膜血管新生为二药配伍治疗缺血性心脏病提供了理论依据.Study of ligustrazine with astragalus armour glycosides compatibility of different concentration of chicken embryos villi allantoic membrane angiogenesis,explore effectively composition effects on ischemic heart disease.Methods: 100 8 d age chicken embryos were randomly divided into 10 groups,including the DMSO blank control group,different concentrations of ligustrazine( 1,2,4 mg / m L) with astragalus glycosides( 0. 2,0. 4,0. 8 mg / m L) two compatibility,a total of nine groups,each group of 10 chicken embryos. Using chicken embryo villus allantois membrane( CAM) method,through the mixed cellulose microporous membrane carrier for the drug to the CAM surface,continue to hatch 3 d observation of blood vessel growth,and new blood vessels were counting. Results:Ligustrazine compatibility and compatibility of astragalus armour glycosides concentration can promote the chicken embryos villi allantois membrane angiogenesis,and has a certain dose-response relationship with concentration. Of ligustrazine 2 mg / m L + astragalus armour glycosides of 0. 4 mg / m L level compared with blank group,blood vessels and secondary vascular tube have obvious difference( P〈0. 001,P〈0. 001). Conclusion: ligustrazine compatibility of astragalus armour glycosides can promote chicken embryo villus urea capsule membrane angiogenesis for effectively compatibility provides a theoretical basis for treatment of ischemic heart disease.
