摘  要:目的:探讨芎芪颗粒抗肾间质纤维化的作用机制。方法:将32只雄性Wistar大鼠随机分入假手术组、UUO模型组、蒙诺治疗组和芎芪颗粒治疗组,采用相应处理后取肾脏组织,以Western Blot检测HGF蛋白表达,以免疫组织化学法检测MMP-3和TIMP-1。结果:与假手术组比较,UUO模型组的肾脏组织HGF蛋白相对表达量下降(P<0.01),MMP-3和TIMP-1均升高(P<0.01)。与UUO模型组比较,蒙诺治疗组和芎芪颗粒治疗组的肾脏组织HGF蛋白相对表达量和MMP-3均升高(P<0.01),TIMP-1均下降(P<0.01)。结论:芎芪颗粒可能通过提高HGF的表达及诱增其下游因子MMP-3的表达,抑制TIMP-1的表达,恢复MMP-3/TIMP-1的失衡,来达到抗肾间质纤维化的作用。Objective: To explore the mechanism underlying Xiongqi Granules prevented renal interstitial fibrosis. Methods: Thirty-two male Wistar rats were randomly divided into the sham-operation group, unilateral ureteral obstruction(UUO) group, monopril group and Xiongqi Granules group. The protein expression of hepatocyte growth factor(HGF) was detected with Western Blot, and marix metalloproteinase-3(MMP-3) and tissue inhibitor of metalloproteinase-1(TIMP-1) were detected with immunohistochemical method. Results: Compared with the sham-operation group, the protein expression of HGF in the UUO group was lower(P〈0.01), and the expression of MMP-3 and TIMP-1 were higher(P〈0.01). Compared with the UUO group, the protein expression of HGF and MMP-3 in the monopril and Xiongqi Granules group were higher(P〈0.01), while the expression of TIMP-1 was lower(P〈0.01). Conclusion: Xiongqi Granules may achieve the effect of anti-renal interstitial fibrosis by improving the expression of HGF and its downstream factor MMP-3, as well as inhibiting the expression of TIMP-1, ultimately recovering the imbalance of MMP-3/TIMP-1.
