基于RACE方法的草莓Faapx-c基因克隆、表达及其生物信息学分析

摘　　要：以草莓栽培品种"丰香"为试材,利用RT-PCR和3′-RACE技术克隆胞质抗坏血酸过氧化物酶基因Faapx-c,并对其进行了生物信息学分析。结果表明:Faapx-c基因长1 034bp,所编码的蛋白FaAPX-c是一个结构稳定、亲水性、非分泌型的蛋白质,其二级结构主要以α?螺旋和无规则卷曲2种形式出现,具有可靠的三维结构,属于植物过氧化物酶家族。Faapx-c基因在草莓的根、茎和叶中能被低温诱导,表达量先升高后下降,而在花中的表达随低温时间的延长而消失。该研究为进一步进行Faapx-c基因的功能研究奠定了基础。Taking Fragaria×ananassacv.Toyonaka as material,a cytosolic ascorbate peroxidase gene Faapx-c was cloned by RT-PCR and 3′-RACE technology,and the bioinformatic analysis of Faapx-c was carried out.The results showed that Faapx-c was 1 034 bp in length coding aprotein FaAPX-c,which was a stable,hydrophilicity and non-secreted protein.The secondary structure of FaAPX-c contained mainly α-helix and random coil,with reliable 3Dstructure.And the protein belonging to a member of plant peroxidase superfamily.Expression analysis of Faapx-c gene showed that it was induced by low temperature in roots,stems,leafs,and the expression amounts were increased firstly and then decreased,but gradually disappeared in flowers.This study laid the foundation for further research on the function of Faapx-c gene.
