摘  要:目的建立HPLC法同时测定八正颗粒中栀子苷、大黄素和大黄酚的含量。方法色谱柱:Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm);柱温:25℃;流动相:甲醇-0.2%磷酸溶液,梯度洗脱;流速:1.0 ml·min^-1;检测波长:254nm;检测时间:30 min。结果栀子苷、大黄素和大黄酚的线性范围为分别为10.6~212、0.515~10.3、1.01~20.2μg·mL^-1。平均回收率分别为100.7%、101.4%、100.2%,RSD分别为0.82%、0.52%、1.29%(n=6)。结论本文方法操作简便、准确度高,能缩短八正颗粒样品的检测时长,提高栀子苷、大黄素和大黄酚含量测定结果的准确性。Objective To establish an HPLC method for determining the content of gardenoside,emodin and chrysophanol in Bazheng granules. Methods Agilent Eclipse XDB-C18( 4. 6 mm × 250 mm,5 μm) chromatographic column was used. The column temperature was 25 ℃. The mobile phase was methanol-0. 2% phosphoric acid with gradient elution. The flow rate was 1. 0 ml·min^-1and the detection wavelength was set at 254 nm. Results The linear range of gardenoside,emodin and chrysophanol was 10. 6- 212,0. 515- 10. 3 and 1. 01- 20. 2μg·mL^-1respectively. The average recovery was 100. 7%,101. 4% and 100. 2% respectively,and RSD was0. 82%,0. 52% and 1. 29%( n = 6) respectively. Conclusion This method is simple,accurate and reliable,which may shorten the detection time of Bazheng granules and improve the determination accuracy of gardenoside,emodin and chrysophanol.
