摘  要:目的探究不同剂量紫外线照射对人脐静脉血管内皮细胞(human umbilical vein endothelial cell,HUVEC)的细胞形态、细胞凋亡率、活性氧(reactive oxygen species,ROS)、血管内皮生长因子(vascular endothelial growth factor,VEGF)和碱性成纤维生长因子(basic fibroblast growth factor,bFGF)的影响。方法实验分正常组和3个不同剂量辐照组,辐照组分别采用10J·m^(-2)、20J·m^(-2)、40J·m^(-2)紫外线照射血管内皮细胞,通过流式细胞术(flow cytometry,FCM)检测各组细胞凋亡率,荧光试剂盒检测ROS的水平和酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)测定VEGF和bFGF的含量。结果与正常组相比,细胞经不同剂量紫外照射24 h后,细胞凋亡率随着照射剂量的增加而升高;ROS、VEGF、bFGF的含量随着照射剂量的增加而升高。结论紫外照射对血管内皮细胞有一定程度的损伤效应,可能与诱导细胞凋亡,释放ROS和激活VEGF和bFGF有关。Objective To investigate the cell morphology, cell apoptosis rate, reactive oxygen species(ROS) vascu- lar endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in human umbilical vein endothelial cell (HUVEC) with different dose of ultraviolet (UV) irradiation. Methods HUVEC cells were irradiated with different dose of UV(0.10. 20. 40 J. m^-2), and cell apoptosis rate was detected by the flow cytometry (FCM), the level of ROS was measured by immunofluorescence kit, and the level of VEGF and bFGF were measured by enzyme-linked immunosorbent assay(ELISA). Results Compared with the control group, the apoptosis rates were increased; the level of ROS were in- creased; the secretion level of VEGF and bFGF were increased by the increase dose of UV irradiation. Conclusion HU- VEC cells are damaged with UV irradiation, by induced cell apoptosis, released ROS, activated VEGF-bFGF.
