摘  要:目的采用超快速高效液相色谱串联四级飞行时间质谱,对金银花、山银花的整体化学成分进行鉴别,并比较不同品种间化学成分的差异。方法采用Phenomenex C_(18)色谱柱(2.1 mm×100 mm,2.6μm),以含0.1%甲酸的乙腈和0.1%甲酸溶液为流动相梯度洗脱,流速0.3 m L·min^(-1);采用配有ESI电喷雾源的高分辨三重四极杆飞行时间质谱,分别在正、负离子模式下进行检测。结果根据正负模式下获得的一级和二级质谱数据,通过对照品对照、准确分子量、裂解碎片及文献,在所有样品中共确证和指证到59个化合物,包括5种生物碱类、14种有机酸类及其衍生物、13种黄酮类、12种环烯醚萜苷类和15种皂苷类化合物。其中24种化合物在品种之间有差异:山银花中含较多的有机酸和皂苷类成分,而金银花中环烯醚萜及黄酮种类较丰富。山银花的4个品种之间,华南忍冬、红腺忍冬和灰毡毛忍冬成分较为相似,而黄褐毛忍冬则有别于其他。结论本实验系统研究了金银花、山银花的化学成分,为其品种鉴定及质量控制提供了依据。Objective To systematically identify the chemical constituents and compare the differences between Lonicerae Japonicae Flos(LJF) and Lonicerae Flos(LF) by UFLC-Triple-Q-TOF-MS/MS. Methods Phenomenex C_(18) column(2.1 mm×100 mm, 2.6 μm) was used with acetonitrile(containing 0.1% formic acid) and 0.1% formic acid solution as the mobile phase in gradient elution. The flow rate was 0.3 mL·min^(-1). The detector was high-resolution quadrupole time of flight mass spectrometry with ESI ion source, tested in both positive and negative model. Results Based on the analysis of mass spectrometry and certifi cation of reference substances, 59 compounds were confirmed, including 5 alkaloids, 14 organic acids, 13 flavonoids, 12 iridoid glycosides and 15 saponins. Twenty-four compounds showed different distribution patterns in LJF an LF. LF had more organic acids and saponins while LJF was more abundant in iridoid glycosides and flavonoids. Among the 4 LF species, Lonicera confusa DC., Lonicera macranthoides Hand.-Mazz. and Lonicera hypoglauca Miq. had similar chemical constituents, different from Lonicera fulvotomentosa Hsu et S.C.Cheng. Conclusion The chemical constituents of LJF and LF are sudied systematically, which provides scientific foundation for species identification and quality control.
