摘 要:利用拟南芥研究木材发育是一种简便而快捷的研究方法。本研究利用前期拟南芥ATH1全基因芯片对毛白杨次生维管系统再生过程中的基因表达分析结果,选择表达差异较大的拟南芥同源基因,由国际拟南芥突变体中心获得拟南芥突变系种子,将其播种进行纯杂合检测以及表型变化研究。研究表明,获得的拟南芥PENNYWISE(PNY)基因突变系均为纯合系,该突变系表型存在明显变化,植株矮化,分枝显著多于野生型分枝。通过冰冻切片进行解剖结构变化分析,发现茎基部次生木质部较野生型发达,细胞排列紧密,韧皮部向外扩增,发育较快,皮层增厚,皮层细胞变大。由此可推测PNY基因可能与维管系统发育相关。本研究将草本模式植物拟南芥与木材发育结合开展研究,可简便快速鉴定相关基因功能,对于木材发育研究具有重要意义。Arabidopsis thaliana has been used to study wood development, which is a kind of simple and efficient research method. This study used gene expression analysis results in the process of regeneration of Chinese White Poplar secondary vascular system by Arabidopsis ATH1 gene chip in the past research work. Arabidopsis homologous genes which have more expression differences were se- lected and Arabidopsis mutant seeds were obtained from the Arabidopsis Biological Resource Center. Sowing these seeds and detecting the homo - or - heterozygous state of these mutants. Phenotypic changes would be researched in this work. The results showed that Arabidopsis mutant pny was homozygote. The mutant phenotype changed obviously. It expressed dwarfing and its branches number was significantly more than the wild type. With the anatomy structure analysis by frozen section, we found that secondary xylem of stem base developed better than wild type. Xylem cells are closer. Phloem part outward expanded and rapidly developed. Cortex became thicken and cells bigger. So presumably PNY gene may be associated with vascular system development. This study combined herb model plant Arabidopsis with woody development for researching. It could be simple and rapid identifying the genes function related to wood formation. That means important research significance for wood development.
