摘 要:目的:通过观察芪桂益脉灵(QGYML)对急性心肌缺血再灌注损伤大鼠血清基质金属蛋白酶-9(MMP-9)表达的影响,探讨该药对心肌缺血再灌注损伤的作用机制。方法:取雄性SD大鼠50只,随机分为5组,正常对照组、假手术组、模型组、芪桂益脉灵高剂量组(1.2 g/kg)和低剂量组(0.6 g/kg)。结扎大鼠冠状动脉左前降支制备急性心肌缺血再灌注损伤模型,造模成功后,分别给予相应处理7 d。采用双抗体夹心酶联免疫法测定血清MMP-9的表达。结果:正常组、假手术组血清MMP-9无明显变化,模型组血清MMP-9水平明显升高,QGYML低剂量组、高剂量组血清MMP-9水平明显降低,与模型组比较均有统计学意义(P<0.05)。结论:QGYML能够通过减少MMP-9的分泌,减轻大鼠心肌缺血再灌注损伤。Objective: To explore the mechanism of protective effect of QGYML on myocardial ischemia-reperfusion injury by observing the effect of QGYML on matrix metalloproteinases-9( MMP-9) expression in rats with ischemia-reperfusion injury. Methods: 50 male SD rats were randomly divided into 5 groups,normal,sham operation,model,QGYML high dose( 1. 2 g / kg) and QGYML low dose( 0. 6 g / kg) groups,models were caused by ligation of left anterior descending branch of coronary artery. After the models were prepared successfully,each group was given the corresponding medicine or saline for 7 days,then the level of serum MMP- 9 was determined by double antibody sandwich method enzyme- linked immunoassay. Results: Serum MMP- 9 level of the normal and sham operation groups had no obvious change,serum MMP- 9 level of the model group increased significantly to compare with normal level,serum MMP- 9 level of QGYML low dose and high dose groups decreased significantly compared with model group,the difference was statistical significance. Conclusion: QGYML can alleviate myocardial ischemia- reperfusion injury in the rat by reducing the secretion of MMP- 9.
