苦参碱联合胸腺五肽对胃癌放化疗大鼠免疫功能的影响

摘　　要：目的探讨苦参碱联合胸腺五肽对胃癌行放化疗大鼠免疫功能的影响。方法将130只成功造模的胃癌大鼠随机分为对照组和观察组,每组65只。2组大鼠均进行放疗(10~12 Gy)、化疗(顺铂^+5-氟尿嘧啶),并同时给予胸腺五肽腹腔注射隔日1次,观察组大鼠在上述基础上给予苦参碱腹腔注射,连续15 d。观察2组大鼠干预前术后免疫功能的变化情况。结果 2组大鼠干预后血清CD3^+、CD4^+、自然杀伤细胞(NK)、Ig G、Ig M、Ig A水平与CD4^+/CD8^+均较干预前明显降低(P均<0.05),CD8^+较干预前明显升高(P均<0.05),但观察组各指标水平均明显优于对照组(P均<0.05);2组大鼠干预后IL-2水平均明显低于干预前(P均<0.05),IL-6水平明显高于干预前(P均<0.05),但观察组变化较对照组更明显(P均<0.05)。结论苦参碱联合胸腺五肽可明显改善行放化疗胃癌大鼠的免疫功能,为临床实践提供了一定的理论基础。Objective It is to investigate the effects of Matrine combined with Thymopeptin on immune function of rats with gastric cancer treated by chemoradiotherapy. Methods 130 rats which were successfully modelled gastric cancer were randomly divided into two groups： observation group（ n = 65） and the control group（ n = 65）. All the rats were given radiation treatment（ 10- 12 Gy）,chemotherapy（ cis- platinum ~＋ 5- fluorouracil）,at the same time given Thymopenptin intraperitoneal injection once every two days. Whereas the rats in the observation group were given Matrine on this basic,the course of treatment were 15 d. The index of immue function in the two groups of rats were compared before and after treatment. Results After treatment,the levels of CD3~＋,CD4~＋,NK,Ig G,Ig M,Ig A,IL- 2 and CD4~＋/ CD8~＋were markedly lower（ all P 0. 05） but the levels of CD8~＋and IL- 6 were markedly higher than before treatment in two groups（ all P 0. 05）; the level of CD3~＋,CD4~＋,NK,Ig G,Ig M,Ig A,IL- 2 and CD4~＋/ CD8~＋of the observation group were significantly higher but the level of CD8~＋and IL- 6 were significantly lower than those of the control group（ all P 0. 05）. Conclusion Matrine combined with Thymopeptin can improve the immune function in rats with gastric cancer treated by chemoradiotherapy,and provide certain theoretical basis for clinical practice.
