摘 要:采用HPLC对发酵前后的绞股蓝皂苷(Gypenoside)进行了分析,选用75只健康小鼠,随机分为阳性对照组、阴性对照组、低剂量试验组、中剂量试验组、高剂量试验组共5个处理组,每组5只小鼠,3个重复。阴性对照组每日灌服0.5 m L生理盐水,阳性对照组每日灌服0.5 m L未发酵的绞股蓝皂苷溶液,3个试验组分别每日灌服发酵的绞股蓝皂苷溶液0.2、0.5、0.8 m L,连续灌服10 d。结果表明,HPLC分析显示,经枯草芽孢杆菌发酵后的绞股蓝皂苷成分发生了明显变化,且与对照组相比,经过发酵的绞股蓝皂苷能够显著提高小鼠的脾脏指数和胸腺指数(P<0.05),同时还能显著提高小鼠血清中Ig G和Ig A的含量(P<0.05),对小鼠血清中IL-2的产生也有一定的促进作用。说明枯草芽孢杆菌对绞股蓝皂苷具有一定的生物转化作用,且经微生物转化后的绞股蓝皂苷具有促进小鼠免疫器官发育和增强体液免疫的功效。In this study the changes between unfermented and fermented Gypenosides were analyzed and treated to 75 healthy mice which allocated to 5 groups,such as negative control group,positive control group and 3 treatments groups,and administrated with 0.5 m L physiological saline,0.5 m L unfermented Gypenosides,and different dose(0.2,0.5,0.8 m L) of fermented Gypenosides for 10 days, respectively. The HPLC analysis results showed that the components of Gypenosides presented significantly change after fermented by Bacillus subtilis. And compared with the control group,the spleen index and thymus index in treatments groups were significantly increased(P〈0.05). The contents of Ig A and Ig G in serum were significantly improved(P〈0.05),and the production of IL-2 in serum was also promoted in the three treatment groups.These results indicated that Bacillus subtilis had certain effects on the biotransformation of Gypenosides,and the fermented Gypenosides could enhance the immune organ development and the humoral immunity of mice.
