摘  要:基于植物组织培养技术,以层积处理西洋参种子为材料,研究植物生长物质对其和离体种胚休眠的解除作用。结果表明:(1)酒精10 s、次氯酸钠10 min处理是试验研究中西洋参种子消毒的最佳方法;(2)植物生长物质GA_3、ZT、IAA、NAA对西洋参种子的休眠解除要比其离体种胚困难;(3)GA_3是西洋参种胚休眠解除的有效植物生长物质,而其他植物生长物质ZT、IAA、NAA主要起到壮苗作用;(4)培养基MS+2I+5G是试验研究中筛选出适合种胚萌发生长的最佳培养基,可用于无菌苗培养;(5)相同的植物生长物质作用下,分化发育较好的种胚(裂口种子种胚)更易被解除休眠。American ginseng seeds after stratification were used to investigate the effects of plant growth substances on dormancy breaking of them and their embryos in vitro based on the plant tissue culture. The results showed that:( 1) the best disinfection method for American ginseng seeds was alcohol 10 s,sodium hypochlorite 10 min in this experiment.( 2) dormancy breaking of american ginseng seeds were more difficulty than their embryos in vitro by plant growth substances including GA_3,ZT,IAA,and NAA.( 3) GA_3 was the effective plant growth substance for dormancy breaking of embryos in vitro,other plant growth substances,including ZT,IAA,and NAA were minor ones.( 4) MS + 2I + 5G was the best culture medium suitable for Germination and growth of American ginseng embryos in vitro to develop plantlets.( 5) the dormancy of better developed embryos was more likely to be breaked broken by the same plant growth substances.
