摘  要:目的利用正交设计法对岗梅总皂苷的提取工艺进行研究。方法以齐墩果酸为对照,香草醛-高氯酸法显色,用紫外-可见分光光度法在550nm处测定岗梅总皂苷的含量。采用L9(34)正交试验设计,考察乙醇浓度、料液比、提取时间、提取次数4个因素对岗梅总皂苷提取的影响,以岗梅中总皂苷的含量为考察指标。结果最佳的提取工艺为40%乙醇,乙醇用量为90倍量,回流提取3次,1.5h·次^(-1)。结论该含量测定方法简便、准确、重复性好,提取工艺经济、合理,值得进一步推广,对工艺开发有较好的参考价值。OBJECTIVE To optimize extraction technology of total saponins in ilex asprela. METHODS The ultraviolet spectrophotometry method was used to measure the total saponins content in ethanol extract of ilex asprela at the maximum absorption wave-length of 550 nm with different polar solvent extraction,with vanillic aldehyde as color reagent and the oleanolic acid as standard substance. On the basis of the single factor test,the effects of ethanol concentration,extraction time,solid / liquid ratio and extraction times on the total saponins extraction process were studied by the by L9( 34) orthogonal experimental design. RESULTS The optimum extraction conditions were as follows: 40% of ethanol solution for 1. 5h,solid / liquid ratio being 1: 90( m / v) and extracting times being three times. CONCLUSION The experimental reproducibility of the process,the process of further development of a better reference value.
