摘  要:为了寻找影响猪产仔数的功能基因,本研究选取5个候选SNP位点CASI0003452、DIAS0001380、ALGA0071919、MARC0052565和ALGA0045832,采用等位基因特异PCR方法对候选位点的多态性变化进行群体研究。结果表明:香猪群体中C^(452)位点以BB基因型为主,低产群的B等位基因频率明显高于高产群(p<0.05),两个香猪群体的B等位基因频率明显高于大白猪(p<0.05)。D^(380)位点,两个香猪群的A等位基因频率低于于大白猪,且与乳头数呈弱的正相关;香猪和大白猪群体中A^(919)和M^(565)位点在香猪和大白猪群体中的基因型均为CC型,没有多态性;A^(832)位点两个香猪群A等位基因频率明显低于大白猪(p<0.05)。经测序证实D^(380)位点处于C1GALT1基因外显子1中,属于同义密码子突变(GCC→GCU),存在密码子偏好性,可能影响香猪C1GALT1基因的表达,可能与香猪的性早熟有关。C^(452)位于PDIA4基因内含子4中,已知PDIA4基因直接影响繁殖的主效基因ER的结构和功能,推测C^(452)位点的多态性有可能影响PDIA4蛋白的表达,进而干扰卵母细胞成熟、受精及胚胎的早期发育等生理过程。To identify single nucleotide polymorphisms(SNPs) and candidate genes affecting reproduction traits of Xiang pig, allele-specific PCR method was used to study the polymorphism of five candidate SNP sites,CASI0003452, DIAS0001380, ALGA0071919, MARC0052565, ALGA0045832. In large pig populations, the allele B frequency at site C(452) in Xiang pig with low litter size was significantly higher than that in Xiang pig with high litter size(p〈0.05), and both of them was higher than that in Yorkshire pig. Allele A frequency of D(380) locus in Xiang pig was lower than that in Yorkshire pig, which was weakly correlated with the nipple number of Xiang pig. Both of A(919) and M(565) sites presented homozygous genotypes in Xiang pig and Yorkshire pig populations without polymorphism. Allele A frequency of A(832) locus in Xiang pig was lower than that in Yorkshire pig.Furthermore, variation of C/T at D(380) locus was confirmed located in exon 1 of C1GALT1(C1GALT1-specific chaperone 1-like) gene, which was characterized as synonymous mutation. But we did find that GCC was the preference codon for C1GALT1 gene with higher frequencies in Yorkshire pig population. It was proposed that the polymorphism at site D(380) might related with the C1GALT1 gene expression and the precocious puberty in Xiangpig. C(452) locus located at intron 4 of PDIA4(protein disulfide isomerase 4) gene. PDIA4 is reported to play an important role in the structure and function of estrogen receptor(ER). It was inferred that the polymorphism at site C(452) might be related with the PDIA4 gene expression, which might further disturb the processes of oocyte maturation,fertilization and development of embryo in Xiang pig.
