摘 要:目的:探讨通络益智散对脑缺血损伤大鼠海马亚区c-Fos蛋白表达的影响。方法:动物随机分为模型组、通络益智散组和尼莫地平组,尼莫地平组、通络益智散组分别灌胃给予尼莫地平0.001 g·kg-1,通络益智散1.57 g·kg-1,模型组给予蒸馏水。预防性灌胃给药7 d后,采用双侧颈总动脉缺血再灌注法制造大鼠脑缺血损伤模型,缺血再灌后2 h、4 h分别免疫组化法检测各组大鼠海马亚区c-Fos蛋白表达情况,并进行组间均数比较。结果:各组海马CA1、CA2、CA3、齿状回、下托区均可见c-Fos的阳性表达。与模型组比较,通络益智散、尼莫地平组海马各亚区c-Fos蛋白表达数量在造模后2h、4h明显降低(P<0.01)。通络益智散组与尼莫地平组比较无明显差异(P>0.05)。各组c-Fos蛋白数量在脑缺血损伤后2 h均达到高峰,4 h出现下降,对比有明显差异(P<0.01)。结论:通络益智散能够抑制脑缺血损伤c-Fos蛋白表达,具有神经保护作用。Objective: The present study was undertaken to investigate the effects of Tongluo Yizhi Powder( TLP) on c-Fos expression in the subregion of hippocampus in acute cerebral ischemia / reperfusion( I / R) rats. Methods: Sprague– Dawley rats were randomly divided into I/R model group,nimodipine group and TLP group. The rats were respectively administrated with TLP or nimodipine for 7 days after the animals were acclimated to the intragastic administration. TLP dissolved in distilled water was dosed at 15. 7 g kg-1per rat and nimodipine dosed at 0. 001 g·kg^-1. The I / R model group was treated with distilled water. The acute cerebral I / R models were established by bilateral carotid artery occlusion. The c-Fos expression was analyzed by ex-vivo Fos immunohistochemistry at 2 h and 4 h after the I / R operation.Results: Moderate to dense c-Fos was expressed in CA1,CA2,CA3,dentate gyrus and subiculum in all rats after the I / R operation. In comparison with the vehicle group,TLP significantly reduced the numbers of c-Fos in CA1,CA2,CA3,dentate gyrus and subiculum( P〈0. 01). There was no significant difference between TLP and nimodipine( P〉0. 05).Moreover,analysis of c-Fos amounts at 2 h and 4 h after I / R operation showed that the expression of c-Fos reached to the peak during 2 h after I / R( P〈0. 01). Conclusion: TLP significantly suppressed the expression of c-Fos in subregion of hippocampus induced by the I / R injury and might play an important role in the brain protection.
