摘 要:目的观察藏药六味木香丸对溃疡大鼠血清中胃泌素(GAS)、环氧合酶-2(COX-2)、NO、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)及丙二醛(MDA)含量的影响,分析其抗溃疡机制。方法通过幽门结扎法建立大鼠胃溃疡模型,采用酶联免疫双抗体夹心法测定GAS、COX-2的含量、采用分光光度法测定NO、NOS、SOD及MDA的含量。结果与模型组比较,六味木香丸高、中剂量均能明显降低大鼠血清中GAS的含量;高剂量能明显抑制血清中COX-2、NO的含量;低剂量能明显升高血清中SOD的水平;六味木香丸各剂量对NOS和MDA的影响均不大。结论六味木香丸抗溃疡的机制之一可能是抑制GAS的产生,从而减少胃酸的分泌;其抗炎机制可能是减少COX-2的产生,升高NO、SOD的含量,从而达到抗氧化、清除自由基的作用。OBJECTIVE To observe the effects of Liuwei Muxiang pills(LWMXP) on gastric secretin(GAS),cyclooxygenase-2(COX-2),nitric oxide(NO),nitric oxide synthase(NOS),superoxide dismutase(SOD) and malondialdehyde(MDA) in the serum of rats with gastric ulcer,and to analyze the antiulcer mechanism of LWMXP.METHODS Rat ulcer model was set up by the way of pyloric ligature.The contents of GAS and COX-2 were detected by Double-antibody enzyme-linked immunosorbent assay method,and the contents of NO,NOS,SOD and MDA were detected by Spectrophotometric method.RESULTS Compared with the model group,high and middle dose of LWMXP could significantly reduce the content of GAS in the rat serum.High dose of LWMXP could inhibit the contents of COX-2 and NO in the serum.Low dose of LWMXP could significantly increase the SOD level in the rat serum.But the content of NOS and MDA didn't show any statistical difference in every dose group.CONCLUSION LWMXP had antiulcer effect on the rat,one of the mechanism might be to reduce the secretion of gastric secretin,thus reduce the parietal cells to produce a large amount of hydrochloric acid in stomach.LWMXP had anti-inflammatory effect,which mechanism may be related to reduction of the content of COX-2,and improve the effect of antioxidant,further scavenging free radicals by increased the content of NO and SOD.
