HPLC测定人脑脊液中的万古霉素和去甲万古霉素

摘　　要：目的 采用HPLC法测定人脑脊液中万古霉素和去甲万古霉素的含量。方法 采用Diamonsil C18柱（150 mm×46 mm,5μm）,流动相为甲醇-0.05 mol·L-1磷酸二氢钾缓冲液（p H3）（20∶80）,流速1.0 m L·min-1,柱温30℃,检测波长236 nm,万古霉素和去甲万古霉素互为内标。结果 脑脊液中万古霉素和去甲万古霉素的线性范围分别为0.0977～97.68（r2=0.9995）、0.104～104μg·m L-1（r2=0.9996）,检测限分别为0.0484、0.0524μg·m L-1;方法回收率分别为99.8%～109.0%、99.6～107.0%,绝对回收率为95.6%～100.0%、95.9%～99.6%,批内、批间RSD均≤4.0%;室温下放置8 h、冻融循环3次和冰冻21 d的稳定性均良好。结论 所用方法简便准确,适宜于临床脑脊液中万古霉素和去甲万古霉素的分析。OBJECTIVE To establish an HPLC method for the determination of Vancomycin and Norvancomycin in human cerebrospinal fluid.METHODS Diamonsil C18column（150 mm × 4.6 mm,5 μm） was used in the method.The mobile phase consisted of methanol and 0.05 mol·L-1potassium dihydrogen phosphate（p H3）（20∶80） at the flow rate of 1.0 m L·min-1.The column temperature was maintained at 30 ℃.The detection wavelength was 236 nm.Vancomycin and Norvancomycin were used as the internal standard for each other.RESULTS The good linear relationship were obtained between the concentration of Vancomycin and Norvancomycinwas from 0.0977 to 97.68 μg·m L-1（r2= 0.9995） and 0.104 to 104 μg·m L-1（r2= 0.9996）.The limit of detection for the analysis of Vancomycin and Norvancomycin were 0.0484 μg·m L-1and 0.0524 μg·m L-1,respectively.The recoveries of method were 99.8%-109.0% and 99.6%-107.0%,respectively.The absolute recovery rates were 95.6%-100.0% and 95.9%-99.6%,respectively.The intra-day and inter-day RSD were all less than 4.0%.Quality control samples of Vancomycin and Norvancomycin in the human cerebrospinal fluid were all stable at the conditions of maintaining at room temperature for 8 h,after freeze-thaw cycle and keeping at-80 ℃ for 21 days.CONCLUSION The method is simple,accurate,and suitable for the analysis of Vancomycin and Norvancomycin in clinical human cerebrospinal fluid.
