熊果酸对糖尿病小鼠视网膜病变的预防作用及其机制探究

摘　　要：目的:研究熊果酸通过调节胰岛素敏感性、葡萄糖转运、血管新生、炎症反应来预防糖尿病小鼠视网膜病变。方法:选择雄性C57BL/6小鼠作为实验动物,随机分为对照组(N组)、模型组(D组)以及干预组(D+UA组),D组和D+UA组采用STZ腹腔注射的方式建立糖尿病模型,D+UA组给予熊果酸灌胃,测定3组小鼠的胰岛素敏感性、视网膜内糖代谢情况以及视网膜组织中GLUTs、HIF-1α/VEGF/VEGFR2通路和IKKβ/IKBα/NF-kB通路的表达量。结果:D组的AUC显著低于N组,HOMA-IR、视网膜组织含糖量以及GLUT-1、GLUT-3、HIF-1α、VEGF、VEGFR2、IKKβ、IKBα、NF-kB、TNF-α、ICAM-1、VCAM-1、E-selectin的含量显著高于N组;D+UA组的AUC显著高于D组,HOMA-IR、视网膜组织含糖量以及GLUT-1、GLUT-3、HIF-1α、VEGF、VEGFR2、IKKβ、IKBα、NF-kB、TNF-α、ICAM-1、VCAM-1、E-selectin的含量显著低于D组。结论:熊果酸能够增加胰岛素敏感性、降低视网膜内含糖量、抑制视网膜组织中的血管新生过程和炎症反应程度,对糖尿病小鼠视网膜病变具有预防作用。Objective:To study the preventive effects of ursolic acid on retinopathy in diabetic mice through adjusting insulin sensitivity,glucose transport,angiogenesis and inflammation.Methods:Male C57BL/6 mice were selected as experimental animals and randomly divided into control group(N group),model group(D group)and intervention group(D+UA group).Diabetes models was established in group D and group D+UA through intraperitoneal injection of STZ.Group D+UA received intragastric administration of ursolic acid,and then insulin sensitivity,glucose metabolism in retina as well as the expression levels of GLUTs,HIF-1α/VEGF/VEGFR2 pathway and IKKβ/IKBα/NF-kB pathway in retina tissue of three groups were detected.Results:AUC of group D was significantly lower than that of N group,and HOMA-IR,sugar content in retina tissue as well as GLUT-1,GLUT-3,HIF-1α,VEGF,VEGFR2,IKKβ,IKBα,NF-kB,TNF-α,ICAM-1,VCAM-1and E-selectin levels were significantly higher than those of N group;AUC of group D+UA was significantly higher than that of group D,and HOMA-IR,sugar content in retina tissue as well as GLUT-1,GLUT-3,HIF-1α,VEGF,VEGFR2,IKKβ,IKBα,NF-kB,TNF-α,ICAM-1,VCAM-1and E-selectin levels were significantly lower than those of group D.Conclusion:Ursolicacid can increase insulin sensitivity,reduce sugar content in retina tissue and inhibit angiogenesis and inflammation degree in retina tissue,and has preventive effect on retinopathy in diabetic mice.
