摘 要:目的:通过比较猪、牛、羊胆汁中胆酸类成分和对发热小鼠清热作用的差异,为胆南星炮制辅料的筛选提供参考依据。方法:采用HPLC-CAD法检测三种胆汁中胆酸类成分(胆酸、去氧胆酸、猪去氧胆酸、鹅去氧胆酸、牛磺胆酸钠)。色谱条件为Waters C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为0.5%甲酸(A)-乙腈(B),梯度洗脱,体积流量:1 mL·min^(- 1);进样量:20μL,Corona参数:氮气压力为35 psi(241.3 Kpa),Filter:high,Range 200 p A。以干酵母制备小鼠发热模型,观察猪、牛、羊胆汁对发热小鼠体温的影响。结果:猪、牛、羊胆汁中胆酸类成分存在明显差异,五种胆酸总量分别为1.495 0%、0.354 0%、0.597 6%,猪胆汁中猪去氧胆酸含量最多,为1.128 7%。猪、牛、羊胆汁对发热小鼠均有一定的清热作用,猪胆汁在6 h时表现出清热作用但不持久。牛胆汁在8 h时表现出较明显的清热作用。羊胆汁清热作用最明显且持久。结论:猪、牛、羊胆汁无论胆酸类成分含量还是清热作用均存在明显差异,提示我们应对药典规定的胆南星的三种胆汁进行区分,将其分别发酵制成胆南星后并结合药效评价再进行筛选。Objective:To compare bile acids composition in pig’s bile,bilis bovina,shee Pbile and differences of clearing heat effect in mice,and provide a reference basis for screening out the arisaema cum bile processing adjuvants.Methods:HPLC-CAD method was used to detect the bile acid composition (cholic acid,deoxidationcholic acid,hyodeoxycholic acid,chenodeoxycholic acid,sodium taurocholic acid) in three kinds of bile.The chromatographic condition was following:Waters C18 column (250 mm×4.6 mm,5 μm),0.5% Formic acid water (A)-Acetonitrile (B),gradient elution,Volume flow:1 mL·min^-1,Corona parameters:the nitrogen pressure was 35 psi (241.3 Kpa),the Filter:high,Range 200 pA.The fever model of mice was induce with dry yeast,the effects of clearing the heat on the model among pig’s bile,bilis bovina and shee Pbile were observed and compared.Results:The total amount of the five kinds of bile acids in pig’s bile,bilis bovina,shee Pbile were 1.495 4%,0.354 0% and 1.495 4%.pig’s bile contains the most amount of hyodeoxycholic acid,was 1.128 7%.The 3 kinds of biles all showed clearing heat function.The pig’s bile showed clearing heat effect at 6 h but not persistent.Bilis bovina showed a significant effect of clearing heat at 8 h.Shee Pbile could clear heat significant and persistent.Conclusion:Both the contents of five kinds of bile acids and the effects of clearing heat are different among the pig’s bile,bilis bovina,shee Pbile,In pharmacopoeia we distinguish the Pinellia Cum Bile with three kinds of bile,then we use pharmacodynamic evaluation to filt the which bile fermente Arisaema Cum Bile is best.
