摘  要:目的:研究去势以及去势后雄激素补充治疗对快速老化小鼠(SAMP8)海马神经元突触蛋白的快速影响.方法:7月龄健康雄性SAMP8小鼠,随机分为假手术对照组(对照组)、去势组、去势+睾酮补充治疗组(T组)和去势+双氢睾酮补充治疗组(DHT组).通过免疫组织化学显色和免疫印迹研究去势及雄激素补充治疗对其海马突触蛋白SNAP25、Syt1、SYN、PSD95、NR1和Drebrin表达的快速影响.结果:免疫组织化学显色结果显示,与SAMP8小鼠假手术组相比,去势组CA1区和CA3区各突触蛋白的OD值均明显降低.T组和DHT组CA1区和CA3区各突触蛋白的OD值较去势组明显增加.免疫印迹结果显示,与SAMP8小鼠假手术组相比,去势组海马结构各突触蛋白条带光密度的相对值均明显降低.T组和DHT组海马结构各突触蛋白条带光密度的相对值较去势组明显增加.结论:SAMP8小鼠去势后,雄激素缺乏导致海马结构内突触蛋白表达减少;去势后雄激素补充治疗能快速增加SAMP8小鼠海马结构内突触蛋白的表达.Objective To demonstrate the rapid effects of androgen on synaptic proteins of hippocampus in male senescence-accelerated-prone mouse 8 (SAMP 8 ) mice after castration and androgens administration. Methods: Male 7-month-old SAM P 8 mice were randomly divided into sham group , cast group , testosterone (T) group and dihydrotestosterone (DHT) group. Using immunohistochemical staining and Western blotting , the expression of SNAP25 , Sytl , SYN , PSD95 , NR1 andDrebrin was determined. Results : Immunohistochemical staining showed the OD value of all of the synaptic proteins in theCA1 and CA3 region was significantly lower in catt group than that in the sham group. T and DHT administration enhancedthe level of synaptic proteins rapidly,the OD value was significantly higher compared to cast group. Western blotting analysis showed that the IOD value of all of the synaptic proteins was lower in the catt group compared to sham group. The IOD value was significantly higher after T and DHT administration than that in the catt group. Conclusion : Castration results in a downregulation of the protein levels of SNAP25 , Sytl , SYN , PSD95 , NR1 and Drebrin. T and DHT treatment significantly improves the level of SNAP25 ,Sytl,S YN,P SD95 ,NR1 and Drebrin protein in a relatively short time.
