联合用药对金黄色葡萄球菌生物膜作用的体外研究

摘　　要：目的观察连花清瘟胶囊水溶解物与头孢替唑钠联合用药对金黄色葡萄球菌生物膜的体外破坏作用。方法药物分为头孢替唑钠组、连花清瘟组、联合用药组和空白对照组;微量肉汤稀释法分别测定连花清瘟胶囊水溶解物和头孢替唑钠对金黄色葡萄球菌的最小抑菌浓度;用酶标仪检测金黄色葡萄球菌生物膜内活菌数变化;用扫描电镜观察金黄色葡萄球菌生物膜内细菌受药物作用后的形态和大小。结果连花清瘟组活菌数较空白对照组低(P<0.05);头孢替唑钠组与空白对照组相比差异无统计学意义(P>0.05);联合用药组活菌数低于单独用药组(P<0.05)。扫描电镜显示头孢替唑钠组生物膜中,大量细菌密集在一起,但密度较空白对照组稀疏,细菌形态基本正常,大小基本一致;连花清瘟组生物膜中形态不规则,大小不一致;而联合用药组形态更加不规则,大小更加不一致。结论连花清瘟胶囊水溶解物对金黄色葡萄球菌生物膜具有显著破坏作用,与头孢替唑钠联合应用可起到协同增效的作用。Objective To observe the destructive effect in vitro of water soluble substance of Lianhua Qingwen capsule combined with Ceftezole Sodium on Staphylococcus aureus biofilm.Methods The drugs were divided into Ceftezole Sodium group,water soluble substance of Lianhua Qingwen capsule group,drug combination group and control group.The minimal inhibitory concentration of water soluble substance of Lianhua Qingwen capsule and Ceftezole Sodium against Staphylococcus aureus was detected by a broth microdilution methed.Staphylococcus aureus biofilm was built and a microplate reader was used to evaluate changes in the number of living bacteria.Scanning electron microscopy （SEM） was used to observe the morphology and thickness of bacteria in Staphylococcus aureus biofilm.Results The number of living bacteria in water soluble substance of Lianhua Qingwen capsule group was decreased compared with the control group （P＜0.05）,while it was not significant in Ceftezole Sodium group （P＞0.05）.The number of living bacteria in the drug combination group was decreased compared with either drug alone group （P ＜ 0.05）.The results of SEM showed Staphylococcus aureus biofilm in Ceftezole Sodium group with a large number of bacteria close together,but the density was a little thin compared with the control group,an almost normal bacterial form,and basically the same size.Staphylococcus aureus biofilm in water soluble substance of Lianhua Qingwen capsule group showed bacteria was irregular form,different size.Staphylococcus aureus biofilm in drug combination group showed bacteria was more inconsistent form,the more inconsistent size.Conclusion Water soluble substance of Lianhua Qingwen capsule has a significant destructive effect on Staphylococcus aureus biofilm in vitro.There was a synergetic destructive effect on Staphylococcus aureus biofilm in combination with Ceftezole Sodium.
