摘  要:目的探究人体富血小板凝胶(PRG)对金黄色葡萄球菌的体外抗菌作用;对比观察不同浓度富血小板凝胶对金黄色葡萄球菌的体外抗菌作用。方法分别取20例健康志愿者外周静脉血30 mnl,制备不同浓度富血小板凝胶,血小板浓度依次为1.8×10^(12)/L(PRG_1组)、1.2×10^(12)/L(PRG_2组)、0.6×10^(12)/L(PRG_3组)。通过体外抗菌实验观察不同浓度PRG在甲氧西林敏感金黄色葡萄球菌(MSSA)、耐甲氧西林金黄色葡萄球菌(MRSA)溶液中孵育1、2、4、8、12、24 h后的抗菌效果。结果在孵育开始后,各浓度PRG菌落计数明显少于PBS对照组,PRG对MRSA和MSSA均有明显的抗菌作用(P<0.05),各浓度PRG抗菌作用均在孵育2 h后最明显,随后逐渐下降;同一时间PRG_1、PRG_2和PRG_3的抑菌率依次下降,与其含血小板浓度呈正相关关系,各组间菌落计数差异均有统计学意义(P<0.05)。在孵育12 h后,细菌生长进入稳定期,各组抑菌率明显下降,各组菌落计数差异无统计学意义(P>0.05)。结论在一定时间内,PRG对MSSA和MRSA均有明显的抑制作用,PRG的抗菌作用与血小板浓度呈正相关关系。Objective To explore the antibacterial effects of human platelet-rich gel (PRG) against Staphylococcus aureus in vitro,comparing the antibacterial effect at different concentrations of PRG against Staphylococcus aureus in vitro.Methods 30 ml peripheral venous from 20 healthy volunteers was prepared for platelet-rich gel with different concentrations.The platelet concentration in PRG1 and PRG2 and PRG3 groups was 1.8 × 10^12/L、1.2 × 10^12/L and 0.6 × 10^12/L,respectively.Antibiotic actions were observed after PRG at different concentrations was incubated in the MSSA (Methicillin-sensitive Staphylococcus aureus),MRSA (Methicillin-resistant Staphylococcus aureus) solution for 1,2,4,8,12,24 hours in vitro.Results The colony count of PRG was significantly less than that of the PBS control group at 12 hours incubation.PRG had obviously antibacterial effect on MRSA and MSSA (P 〈 0.05),the antibacterial effects of PRG were most pronounced at 2 hours;At the same time the inhibitory rate of PRG1 and PRG2 and PRG3 groups were decreased in turn,and containing platelet concentrations were positively correlated.The differences of each colony count were statistically significant (P 〈 0.05).After 12 hours of incubation,when the growth of bacteria had entered the stable phase,the inhibitory rate of each group was significantly decreased.There was no significant difference in the colony count (P〉0.05).Conclusion In a certain period of time,PRG has a significant inhibitory effect on MSSA and MRSA,and the antibacterial activity of PRG was positively correlated with the platelet concentration.
