摘  要:目的利用siRNA干扰技术研究胸腺素β_4(Tβ_4)基因沉默对鹿茸间充质干细胞(MSC)增殖的影响。方法鹿茸MCS分为阴性对照组、siRNA-Tβ_4转染组2组。MTT检测MSC增殖的变化,Real-time PCR检测ILK、AKT及Cyclin D1 mRNA表达量差异变化,Western blot检测AKT及P-AKT蛋白含量变化。结果 siRNA-Tβ_4转染组MSC增殖受到显著抑制(P<0.01);siRNA-Tβ_4转染组ILK、Cyclin D1 mRNA表达量下调,AKT mRNA表达量不变;siRNA-Tβ_4转染组总AKT蛋白含量无变化,P-AKT蛋白含量减少。结论 Tβ_4基因沉默能显著抑制MSC增殖,并可能通过下调ILK、P-AKT及细胞周期相关蛋白CyclinD1从而抑制MSC细胞增殖。Objective To investigate the effects of siRNA-mediated thymosin β4 ( Thymosin β4 ,Tβ4 ) silence on proliferation in activated mesenchymal stem cell( mesenchymal stem cell,MSC) and discuss the possible mechanism.Methods MSC cells were divided into 2 groups: negative control group and siRNA-Tβ4 transfection group. The siRNA-Tβ4 was transfected into MSC cells and cell proliferation was measured by MTT,Real-time PCR detection ILK,AKT and Cyclin D1 mRNA expression quantity change,Western blot detection AKT and P-AKT protein content changes. Results MSC proliferation was significantly inhibited in siRNA-Tβ4 transfection group( P〈0.01); Expression of the ILK,Cyclin D1 mRNA of MSC transfected with siRNA-Tβ4 is reduced,AKT quantity remains the same( P〈0.01); The siRNA-Tβ4 transfection group no change in total AKT protein,P-AKT protein content decreased. Conclusion The siRNA-mediated Tβ4 silence significantly inhibits MSC proliferation by up regulating the ILK,p-AKT and Cyclin D1 expression in MSC.
