摘 要:目的:比较3种补肾方对卵巢摘除骨质疏松大鼠Wnt通路β-Catenin m RNA表达的影响。方法:雌性SD大鼠70只,随机分为假手术组、模型组、龟鹿二仙胶组、二仙汤组、补肾活血组、雌二醇组、仙灵骨葆组,每组10只。双侧卵巢摘除术复制骨质疏松模型,术后30天进行相应的给药实验,假手术组和模型组给予灌服等量的生理盐水。12周后测定各组大鼠血清中I型胶原羧基端肽(β-Crosslaps)、I型前胶原氨基端延长肽(P1NP)含量;以双能X线骨密度测定仪测定大鼠股骨的骨密度;用RT-PCR检测右侧股骨髓LRP5、Wnt2、β-Catenin m RNA表达,用Western blot方法检测LRP5、Wnt2、β-Catenin蛋白表达。结果:与sham组比较,模型组股骨骨密度、血液PINP浓度、β-Catenin m RNA、LRP5 m RNA、Wnt2 m RNA表达均显著降低(P<0.01),β-Crosslaps浓度显著增高(P<0.01)。与模型组比较,3组中药组骨密度、血液PINP浓度、LRP5 m RNA、β-Catenin m RNA、Wnt2 m RNA及蛋白表达显著增高(P<0.05),血液β-Crosslaps浓度显著降低(P<0.01),补肾活血组骨密度及β-Catenin m RNA及蛋白表达高于其他两补肾方剂组。结论:补肾方通过Wnt信号通路促进去卵巢大鼠成骨细胞增殖而改善骨质疏松症,补肾活血组方效果更为明显。This study aimed to explore the effects of three differenct prescriptions of tonifying kidney on the expression of β-Catenin m RNA of Wnt signaling pathway in osteoporosis rats. In this experiment, 70 female SD rats with the weight of 250±20 g were randomly divided into seven groups: the sham-operation group(the control group), the model group, the Guilu Erxian glue group, the Erxian decocotion group, the torifying kidney and promoting blood flow treatment group(TCM group), the estradiol treatment group(the E group) and Xianlin Gubao group, with 10 rats for each group. The osteoporosis model was established by bilateral ovariectomy. The control group underwent sham surgery, while the other six groups administered by bilateral ovariectomy. Rats had been given corresponding treatments for 30 days after the operation. And twelve weeks later, the femoral bone mineral density(BMD) was detected by dual-energy X-ray absorptiometry, and plasma type I collagen carboxy-terminal peptide(β-Crosslaps) and amino-terminal propeptide of type I procollagen(PINP), the m RNA and protein expression of Wnt2, LRP5 and β-catenin extracted from the right femoral bone marrow weretested by RT-PCR and western blot, respectively. It was found that compared with the control group, BMD, plasma PINP level and the m RNA and protein expressions of Wnt2, LRP5 and β-Catenin in the model group were decreased significantly(P 〈 0.01), while the β-Crosslaps expression increased(P 〈 0.01). Compared with the model group, BMD, PINP and the m RNA and protein expressions of Wnt2, LRP5 and β-Catenin in the three groups treated by three different tonifying-kidney prescriptions were increased obviously(P 〈 0.05), while the expression of β-Crosslaps attenuated(P 〈 0.01), and the BMD, the m RNA expression of β-Catenin in the TCM group was higher than that in the Guilu Erxian glue group and the Erxian decocotion group.
