摘 要:目的建立树鼩单纯疱疹病毒(Herpes Simplex Virus,HSV)感染模型,为研究HSV潜伏感染机制、病毒与宿主相互作用的表观遗传学机制提供基础数据。方法先在不同细胞系中比较HSV-1 17+毒株的感染效果,然后用HSV-1 17+毒株通过眼膜划痕的方法感染树鼩,观察其发病情况及死亡率;采集树鼩脑组织进行PCR检测,并采血检测HSV-1抗体。结果 HSV-1 17+毒株可感染多种细胞并引起细胞发生病变;树鼩感染死亡率约为33.33%,树鼩脑组织采用PCR均检测到HSV-1基因组;采用ELISA检测树鼩感染HSV-1后血清特异性抗体阳性率偏低,2周和3周时分别为4.55%和5.56%。结论成功建立树鼩HSV-1感染模型,在感染动物组织中检测到HSV-1DNA,部分感染动物血清中检测到HSV-1IgG抗体,为HSV相关研究奠定基础。Objectives To establish latent herpes simplex virus(HSV)infection in a tree shrew model,study the mechanisms of latent infection,and provide basic data on the epigenetic mechanisms of interaction between the virus and host. Methods The effectiveness of infection with the 17+ strain of HSV-1 was examined in different cell lines.Tree shrews were infected with the 17+strain of HSV-1by corneal scarification.The prevalence of the disease was noted and the mortality rate was recorded.Brain tissue was collected from tree shrews and subjected to PCR.Blood was collected from the thigh vein to detect antibodies against HSV-1with ELISA. Results The 17+ strain of HSV-1infected many types of cells and caused a cytopathic effect(CPE).Tree shrews that were infected by corneal scarification had a mortality rate of about 33.33%.PCR detected the HSV-1genome in brain tissue from tree shrews infected with the 17+strain on days 7and 28post-infection.ELISA data indicated that infected tree shrews tested positive for IgG antibodies against HSV-1at a low rate(4.55% 2weeks post-infection and 5.56% 3 weeks post-infection). Conclusion A tree shrew model of HSV-1infection was successfully established.HSV-1DNA was detected in tissue from tree shrews infected with HSV-1,and IgG antibodies against HSV-1were detected in some serum samples.These data have laid the foundation for related research on HSV.
