摘  要:目的优化易变链霉菌Streptomyces mutabilis TRM 45540产放线菌素D的发酵条件,以期提高放线菌素D的产量。方法在单因素试验的基础上通过Plackett-Burman试验设计、最陡爬坡试验以及中心组合试验优化。结果最适培养基为燕麦片30.0 g/L,大豆粉15.0g/L,氯化钠19.0g/L,磷酸氢二钾1.5g/L,七水硫酸镁2.5g/L,碳酸钙1.0g/L;在摇床转速180r/min,初始p H 7.0,装液量150/500m L,接种量4%,温度28℃条件下发酵7d,TRM45540菌株产放线菌素D的产量为679.2mg/L。结论优化后产量比优化前提高了6.5倍,用该菌株生产放线菌素D具有发酵原料经济、发酵时间短、收率高和杂质少的优点,S.mutabilis TRM45540可以作为生产放线菌素D的候选菌株。Objective The fermentation conditions were optimized for Streptomyces mutabilis TRM45540to produce actinomycin D. Methods By single factor experiment, Plackett-Burman test, the steepest ascending experiment as well as central composite design and response surface method. Results The optimized fermentation conditions were as follows: oatmeal 30.0g/L, soybean powder 15.0g/L, sodium chloride 19.0g/L, potassium hydrogen phosphate 1.5g/L, magnesium sulfate heptahydrate 2.5g/L, calcium carbonate 1.0g/L; rotating speed 180r/min, initial pH value 7.0, broth content 150mL/500mL, inoculums size 4%, culture temperature 28℃, and fermentation period 7d. Under these optimum conditions, the yield of actinomycin D was obtained up to 679.2mg/L. Conclusion It was increased by 6.5 times compared with the original value before oPtimization. It takes the advantages of high yield, low impurities, cheap materials and short fermentation time for Streptomyces mutabilis TRM 45540 to produce actinomycin D.
