摘  要:目的观察维生素E、依达拉奉对大鼠转移术后扩张皮瓣氧自由基的清除作用。方法24只成年SD大鼠随机分成对照组、维生素E治疗组、依达拉奉治疗组,每组8只。于各组大鼠背部均埋入30ml软组织扩张器1只,术后定期注水扩张至32ml。再次手术取出软组织扩张器,并于背部扩张区形成蒂部位于尾侧的2.0cm×6.0cm的任意皮瓣。皮瓣形成后即刻,维生素E治疗组、依达拉奉治疗组、对照组大鼠分别经尾静脉注射维生素E注射液(100mg/kg)、依达拉奉注射液(10mg/kg)、生理盐水1ml;术后24h分别取材检测各组大鼠皮瓣的丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、髓过氧化物酶(MPO)活性;并通过苏木素-伊红(HE)染色观察皮瓣组织结构变化及炎性细胞浸润,计数多核炎性细胞数。结果皮瓣的MDA含量、MPO活性、多核炎性细胞数在维生素E治疗组分别为:(9.25±0.33)nmol/mg prot、(0.47±0.05)U/mgprot、(5.36±0.29)个/高倍视野,在依达拉奉治疗组分别为:(6.12±0.53)nmol/mgprot、(0.35±0.03)U/mg prot、(3.71±0.33)个/高倍视野,结果均显著低于IR组[(11.74±0.29)nmol/mg prot、(0.61±0.02)U/mg prot、(7.43±0.62)个/高倍视野,P〈0.05];且依达拉奉治疗组皮瓣的MDA含量、MPO活性、多核炎性细胞数均显著低于维生素E治疗组。皮瓣的SOD活性在维生素E治疗组为(25.36±2.01)U/mg prot,在依达拉奉治疗组为(28.23±1.82)U/mg prot,结果均显著高于IR组[(18.61±1.32)U/mg prot,P〈0.05];且依达拉奉治疗组皮瓣的SOD活性显著高于维生素E治疗组,差异均有统计学意义(P〈0.05)。结论依达拉奉、维生素E均能清除转移后扩张皮瓣组织内的氧自由基并减轻炎性反应,且依达拉奉的作用效果优于维生素E。Objective To study the scavenging activity of vitamin E and edaravone on oxygen free radicals in rats' expanded flaps after transplantation. Methods Twenty - four adult SD rats were random- ly divided into control group, vitamin E treated group and edaravone treated group, eight rats in each group. A soft tissue expander of 30 ml was put into back of every rat in all the three groups. After the oper- ation, normal saline was injected into every soft tissue expanders regularly until the total volume reached to 32 ml. All the soft tissue expanders were removed in the other operation, then formed a flap sized 2 cm × 6 cm in the expanded area of back with the pedicle located at the trailing side. After flaps were formed im- mediately, vitamin E injection ( 100 mg/kg), edaravone injection ( 10 mg/kg) and 1 ml normal saline were respectively injected into rats in vitamin E treated group, edaravone treated group and control group via the tail veins. Samples of flaps were harvested from each group 24 hours after the operation, to detect the content of malondialdehyde ( MDA ), activity of superoxide dismutase ( SOD ) and myeloperoxidase (MPO) ; Observed changes in organizational structure and infiltration of inflaimnatory cells by hematoxylin - eosin staining (HE) staining, counted the number of inflammatory ceils with polynuclear. Results The content of MDA, activity of MPO, count of inflammatory cells with muhicore were respectively (9. 25 ±0. 33) nmol/mg prot, (0. 47 ±0. 05) U/rag prot, (5.36 ±0. 29)/high power objective (HP) in vitamin E treated group and (6. 12±0.53) nmol/mg prot, (0.35 ±0.03) U/rag prot, (3.71 ±0.33)/ HP in edaravone treated group; all of the data were significantly lower than that in IR group correspondingly with the data ( 11.74 ±0. 29) nmol/mg prot, (0. 61 ±0.02) U/mg prot, (7. 43 ±0. 62)/HP(P 〈0. 05) ; and the content of MDA, activity of MPO, count of inflammatory cells with multicore in edaravone treated group wer
