摘 要:目的探讨健脾益肺化痰系列方对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)模型大鼠气道黏液高分泌的影响。方法采用单纯香烟烟熏法建立COPD大鼠模型,60只Wistar雄性大鼠随机分为正常对照组、模型组、健脾益肺化痰方组、健脾益肺方组、化痰方组和羧甲司坦组,每组10只。健脾益肺化痰方组、健脾益肺方组、化痰方组和羧甲司坦组大鼠分别给予相应的药物灌胃治疗,正常组和模型组灌胃等体积的纯净水。末次给药后,禁食12 h后取左肺中叶肺组织,用于实时荧光定量PCR检测及免疫组化法检测肺组织黏蛋白5AC(mucin5AC,MUC5AC)和钙离子激活的氯离子通道1(calcium activated chloride channel,CLCA1)mRNA水平及蛋白表达。结果与正常组相比,模型组大鼠肺组织MUC5AC和CLCA1 mRNA水平及蛋白表达均增多(P<0.01)。与模型组比较,健脾益肺化痰方组、健脾益肺方组、化痰方组的大鼠肺组织MUC5AC和CLCA1 mRNA水平及蛋白表达均减少(P<0.01)。其中,健脾益肺化痰方组大鼠肺组织MUC5AC和CLCA1 mRNA水平及蛋白表达减少最明显。结论健脾益肺化痰系列方能有效抑制COPD模型大鼠气道黏液高分泌,可能是与健脾益肺化痰系列方的"化痰"作用有效降低了肺组织MUC5AC和CLCA1 mRNA水平及蛋白的表达相关。Objective To investigate the influence of Jianpiyifeihuatanfang on airway mucus hypersecretion in rat model of chronic obstructive pulmonary disease( COPD). Methods The 60 male Wistar rats were randomly divided into normal control group,model group,Jianpiyifeihuatan group,Jianpiyifei group,Huatan group,and Carbocisteine group,each group of 10. In addition to the normal control group,the model of COPD rats on other groups was established by tobacco smoke inhalation. Jianpiyifei group,Huatan group,Jianpiyifeihuatan group and Carbocisteine group were given the lavage treatment with corresponding drug,model group and normal control group were given equal volume of pure water. The eating was banned for 12 hours after the last delivery. Then take the left middle lobe of rats.These cases were stained by PCR staining and IHC staining. Last,observe the pathological change of rat lung tissue on each group. Results Compared with normal control group,in the model group the level and expression of MUC5 ACmRNA and CLCA1-mRNA increased( P〈0. 01). Compared with the model group,in the Jianpiyifeihuatan group and Jianpiyifei group and Huatan group the level and expression of MUC5AC-mRNA and CLCA1-mRNA decreased( P〈0. 01). Among three groups,the Jianpiyifeihuatan group is obviously decreased. Conclusion A series of Jianpiyifeihuatanfang can obviously inhibite the COPD rat model high airway mucus secretion,there may be related to decrease the level and expression of MUC5AC-mRNA and CLCA1-mRNA by the "reducing phlegm" of a series of Jianpiyifeihuatanfang.
